I Basic Characteristics

It readily stains with basic dyes and shows a positive Gram reaction due to its thick peptidoglycan cell wall.

This organism has relatively low nutritional requirements and can grow on standard nutrient agar.

The optimal growth temperature is around 37°C, with an optimal pH of approximately 7.4.

Colonies on agar plates are typically round, convex, smooth, moist, and opaque, with colors ranging from white to pale yellow.

Growth in CO₂-enriched environments may enhance metabolic activity and toxin production.

III Pathogenicity and Clinical Significance

Although commonly part of the normal flora of skin and mucosal surfaces, Staphylococcus saprophyticus can cause infection when host defenses are compromised.

It is one of the leading causes of uncomplicated urinary tract infections, particularly in young women.

Clinical conditions include cystitis and pyelonephritis, with symptoms such as dysuria, frequency, and lower abdominal discomfort.

It may also cause wound infections under certain conditions.

Staphylococcus saprophyticus is catalase-positive and typically coagulase-negative, distinguishing it from Staphylococcus aureus.

A key diagnostic feature is its resistance to novobiocin, which differentiates it from other coagulase-negative staphylococci such as Staphylococcus epidermidis.

On blood agar, colonies appear white or grayish without significant hemolysis.
