Where the antibody actually goes
To cover a membrane, the liquid only needs to form a few millimeters of depth over it. In a wide single-well tray, reaching that depth across the whole footprint can take 5–10 mL. Cut that same membrane into strips and incubate each strip in a narrow channel, and 1–2 mL per strip is enough for full, even coverage — a 3–5× reduction in prepared antibody per target.
Strip incubation has a second, bigger payoff: you can probe multiple targets from one gel. Cut the membrane by molecular weight, drop each strip into its own channel with a different primary antibody, and one gel yields data on six proteins — target, loading control, and four more — in a single incubation session.
