I Classification and Distribution

Pediococcus pentosaceus belongs to the genus Pediococcus within the lactic acid bacteria group. It is named for its ability to metabolize pentose sugars such as xylose and arabinose.

It is widely distributed in fermented foods including kimchi, sausages, cheese, and beer, as well as on plant surfaces and within animal gastrointestinal tracts.

The organism is generally considered safe and contributes beneficially to food quality and gut health.

II Growth and Biochemical Characteristics

As a facultative anaerobe, Pediococcus pentosaceus grows well under microaerophilic conditions. It is commonly cultured on MRS agar, forming white, round colonies (1–2 mm) after 48–72 hours.

It is catalase-negative and performs homofermentative metabolism, producing lactic acid from glucose without gas formation.

In addition to pentoses, it can metabolize certain hexoses such as glucose and fructose.

The growth temperature range is 15–45°C, with an optimal range of 30–35°C. It can tolerate up to 6.5% NaCl, making it suitable for food fermentation environments.

Pediococcus pentosaceus has significant applications in food and health industries.

Food fermentation: Used as a starter culture in fermented foods such as sausages, kimchi, and beer. It produces bacteriocins such as pediocin, which inhibit spoilage and pathogenic bacteria.

Food safety enhancement: Helps reduce nitrite levels and improve product safety and shelf life.

Gut health: Modulates intestinal microbiota and suppresses pathogens such as Salmonella and Escherichia coli.

Immune support: Stimulates immune responses, including macrophage activation and IgA secretion.

Culture-based identification is performed on MRS agar, followed by biochemical testing using systems such as API 50 CHL or VITEK.

Advanced identification methods include MALDI-TOF MS and 16S rRNA gene sequencing.

Real-time PCR (qPCR) targeting species-specific genes such as rpoA or ddl provides rapid, sensitive, and specific detection for food, clinical, and environmental samples.

VI Clinical Management and Safety Considerations

In rare infection cases, treatment should focus on controlling infection and removing the source (e.g., infected catheter).

Empirical therapy may include antibiotics effective against Gram-positive bacteria such as vancomycin or linezolid, guided by susceptibility testing.

While generally sensitive to vancomycin, resistance to fluoroquinolones and tetracyclines may occur, requiring monitoring of resistance gene dissemination.
