Description
Biofargo Fluorometer S96 is a compact two-channel fluorescence microplate reader for quantifying DNA, RNA, microRNA and protein. It reads a full 96-well plate in 55 seconds, so a batch that takes most of a morning on a single-sample fluorometer is finished in under a minute of instrument time.
Each well needs only 1–20 µL of sample, made up to 200 µL with the assay working solution in a 0.3 mL 96-well flat-bottom black plate. Blue and red excitation with green and red emission detection covers dsDNA, ssDNA, RNA, microRNA and protein assays on one instrument, and a SiPMT detector with top-reading optics gives a dynamic range of five orders of magnitude.
The instrument is run from PC software that handles protocol creation, well layout, sample entry, result review and Excel export, with a Chinese and English interface. Two-point calibration is used for quantification, so each plate carries its own standards rather than a stored factor.

Protocol Overview
Power on the instrument and allow 15 minutes for the optics to stabilize. Connect it to the PC with the supplied cable and open the control software.
Prepare the assay working solution for the chosen Biofargo assay kit and dispense it into a 0.3 mL 96-well flat-bottom black microplate.
Add the kit standards to their wells, then add 1–20 µL of each sample to its well and bring every well to a total volume of 200 µL.
Mix and incubate as the assay kit specifies, protected from light.
In the software, create or select the protocol for the assay, lay out the standard and sample wells, and enter the sample information.
Load the plate and start the run. A full plate reads in 55 seconds.
Review the standard curve and the results on screen, then export to Excel. Dilute and re-read any sample that falls above the top standard.

Which microplate should I use?

Does it need a computer?

What is the sensitivity and detection range?
Sensitivity is 0.05 ng/µL for dsDNA, 0.1 ng/µL for ssDNA and microRNA, and 0.5 ng/µL for RNA. Detection ranges are 0.01–100 ng/µL for dsDNA HS, 1–1,000 ng/µL for dsDNA BR, 0.25–100 ng/µL for RNA HS and 1–1,000 ng/µL for RNA BR.
How is the instrument calibrated?
Quantification uses two-point calibration with the standards supplied in the assay kit. Run the standards on the same plate as the samples in every batch rather than relying on a stored curve.
Can it read raw fluorescence without a standard curve?
Yes. RFU mode reports fluorescence intensity directly, which is useful for relative measurements and for checking a new assay before building a curve.
How long does it take to stabilize after power-on?
Allow 15 minutes after power-on before the first read.
