Description

Laboratories measuring double-stranded DNA by UV absorbance at 260 nm may encounter interference from RNA, proteins, salts, and detergents. The Biofargo dsDNA BR Assay Kit uses a fluorescence-based method that is highly selective for dsDNA over RNA and is minimally affected by the common contaminants evaluated in the user guide.

Depending on sample volume, the assay measures initial dsDNA concentrations from 0.2 to 1,000 ng/µL, with a total assay range of 2–1,000 ng. Each assay uses 1–20 µL of sample in a final volume of 200 µL. The protocol supports a Qubit fluorometer or a fluorescence microplate reader.

The assay requires a 2-minute incubation at room temperature protected from light. The signal remains stable for up to 3 hours after incubation when protected from light. For research use only; not for diagnostic or therapeutic procedures.

Features

  • Broad-range dsDNA measurement with a total assay range of 2–1,000 ng.

  • Highly selective for double-stranded DNA over RNA.

  • Accepts 1–20 µL of sample in a final assay volume of 200 µL.

  • Protocols for a Qubit fluorometer and a fluorescence microplate reader.

  • A 2-minute incubation at room temperature, protected from light.

  • Fluorescent signal remains stable for up to 3 hours after incubation when protected from light.

Applications

  • Qubit Fluorometer: Quantify dsDNA in clear, thin-wall 0.5 mL PCR tubes using the two supplied standards and the dsDNA Broad Range assay setting.

  • Fluorescence Microplate Reader: Quantify dsDNA in a black-wall, clear-bottom 96-well microplate using a standard curve, 485 nm excitation, and 530 nm emission.

Frequently Asked Questions (FAQs)

What is the detection range of the Biofargo dsDNA BR Assay Kit?

The total assay range is 2–1,000 ng. Depending on sample volume, the initial dsDNA concentration range is 0.2–1,000 ng/µL.

Can the kit be used with a Qubit fluorometer?

Yes. The user guide includes a Qubit workflow using two standards. On a Qubit 4 Fluorometer, select dsDNA and then dsDNA Broad Range.

Can the kit be used with a fluorescence microplate reader?

Yes. The user guide includes a 96-well workflow with 485 nm excitation and 530 nm emission.

How much sample is needed?

Add 1–20 µL of sample and adjust the working solution so that the final volume is 200 µL per tube or well.

How should the kit be stored?

The kit is shipped on ice and should be stored at 2–8 °C upon arrival. Protect the fluorescent dye and freshly prepared working solution from light.
