Product Description

SHENTEK® Residual CV-1 DNA Quantitation Kit is designed to quantitate residual CV-1 host cell DNA across stages of biopharmaceutical production, from in-process samples to final products. The kit uses quantitative PCR (qPCR) to deliver rapid, specific and reliable quantitation at the femtogram (fg) level and includes a CV-1 DNA Control reference standard. • Prepare standard curve fresh using serial dilutions; include at least five concentrations for curve fitting.

• Seal plates with appropriate film, mix and spin briefly before placing on instrument.

• Acceptance criteria: ERC recovery 50%–150%; NTC Ct ≥35 cycles or undetermined (or per lab validation); NCS Ct must be > mean Ct of lowest standard or below validated LOQ.

Q1: What is the assay sensitivity or lowest detectable concentration?
The kit provides a lowest standard of 0.003 pg/µL (3 fg/µL) and is designed to support femtogram-level quantitation. The validated limit of quantitation (LOQ) should be determined by laboratory validation and may be lower or higher depending on sample matrix and extraction recovery.
Q2: How should reagents be stored and handled?
Store kit components at recommended temperatures (major components at -20 °C). Thaw reagents on ice (2–8 °C), vortex and spin briefly before use. Protect light-sensitive components (e.g., qPCR Reaction Buffer) from light. Follow MSDS guidance and use DNase-free consumables.
Q3: What acceptance criteria should I use for controls?
ERC recovery should be between 50% and 150%. NTC should have Ct ≥35 cycles or be undetermined (or per laboratory validation). The NCS Ct should be larger than the mean Ct of the lowest concentration in the standard curve or below the validated LOQ.
Q4: Which instruments are supported and what detector settings are recommended?
