Description

Terminal deoxynucleotidyl transferase (TdT) is a non-template dependent DNA polymerase that catalyzes the addition of dNTPs to the 3' hydroxyl end of oligonucleotides, single-stranded DNA, and double-stranded DNA, with the oligonucleotide needing to contain at least 3 nucleotides. TdT can also act on RNA, but its activity is strictly dependent on the tertiary structure of the receptor RNA's 3' end and the type of nucleotides present. Generally, TdT has lower efficiency when acting on RNA compared to DNA.

Applications

  • Oligonucleotide or DNA 3' hydroxyl end labeling DNA tailing Synthesis of oligonucleotide chains with the same type of deoxyribonucleotides 5'-RACE (Rapid Amplification of cDNA Ends).

  • Enzyme Activity: One unit (U) of TdT activity is defined as the amount required to catalyze the addition of 1 nmol of dNTP to the 3' hydroxyl end of a polymeric nucleotide within 60 minutes at 37°C.

  • Inactivation: TdT can be inactivated by heating at 70°C for 10 minutes or by adding an appropriate amount of EDTA.

  • Inhibitors: TdT activity is inhibited by metal ion chelators, as well as by higher concentrations of ammonium ions, chloride ions, iodide ions, and phosphate ions.
