Post-Thaw NK Cell Viability After Cryopreservation

Cryopreservation performance was evaluated using human NK cells with a CD3-CD56+ proportion greater than 90%. Cells were generated from cord blood-derived mononuclear cells and expanded using an XR NK activation workflow before freezing with XR NKCryo.

Recommended Cryopreservation Workflow

XR NKCryo provides a standardized workflow for laboratories performing human NK cell expansion, freezing and recovery experiments. The recommended process includes cell harvesting, cryoprotectant addition, controlled-rate freezing and transfer to long-term storage.

Step 1: NK Cell Collection

After completion of NK cell culture and expansion, collect NK cells by centrifugation and carefully remove the culture supernatant.

Step 2: Add XR NKCryo Freezing Medium

Slowly add XR NKCryo cryopreservation medium to the cell pellet. Gently resuspend the cells to achieve a uniform suspension.

Recommended final freezing density:

Step 3: Controlled Cooling and Storage

Transfer prepared cryovials into a controlled-rate freezing container and place them at -80°C. For long-term storage, transfer cells to liquid nitrogen storage after approximately 12 hours.

Thaw frozen NK cells using a 37°C water bath or cell recovery instrument.
