Description

Establishing reliable human lung cancer organoid models from primary tumor tissues can be challenging due to variations in tumor samples, limited expansion capacity, and the difficulty of maintaining tumor-specific characteristics during in vitro culture. Researchers often require a standardized culture environment that supports consistent organoid establishment while preserving the biological relevance of patient-derived tumor models.

XR Human Lung Cancer Organoid Culture Kit (XRhLungCO-100) is a specialized lung tumor organoid culture system designed for the generation, expansion, and maintenance of human lung cancer organoids derived from tumor tissues. The kit provides a standardized culture solution consisting of human lung cancer organoid basal medium A and culture supplement B, enabling researchers to prepare a complete organoid culture medium for 3D tumor model development.

This human lung cancer organoid culture kit supports patient-derived lung cancer organoid research workflows, including tumor biology studies, disease modeling, cancer drug screening, and precision oncology research. By providing a defined culture environment, XR organoid medium helps researchers establish reproducible 3D lung cancer models for advanced cancer research applications.

Human Lung Cancer Organoid Culture Workflow

The establishment of patient-derived lung cancer organoids typically requires efficient tissue processing, controlled dissociation, extracellular matrix embedding, and specialized organoid culture conditions. XR Human Lung Cancer Organoid Culture Kit is designed to support a standardized workflow from primary tumor tissue preparation to 3D organoid expansion.

Step 1: Tumor Tissue Collection and Preservation

Freshly isolated lung cancer tissue samples are temporarily preserved in pre-cooled primary tissue storage solution and maintained under refrigerated conditions before processing.

Step 2: Tissue Washing and Mechanical Processing

Under sterile conditions, tumor tissues are washed repeatedly with pre-cooled PBS to remove residual impurities. Clean tissues are then transferred into sterile tubes and mechanically cut into small tissue fragments approximately 1–3 mm³ in size.

Step 3: Tissue Dissociation and Cell Preparation

Prepared tissue fragments are treated with primary tissue digestion solution at 37°C for approximately 15–45 minutes depending on tissue source and size. Following digestion, the cell suspension is filtered through a 100 μm cell strainer to remove undigested tissue fragments.

Step 4: Organoid Embedding and 3D Culture Initiation

Collected cells are mixed with extracellular matrix material at an optimized density range of 1×10⁵–1×10⁶ cells/mL. Cell-matrix droplets are placed into culture plates and incubated at 37°C with 5% CO₂ until matrix solidification.

Step 5: Lung Cancer Organoid Expansion

After matrix solidification, complete lung cancer organoid culture medium prepared from basal medium A and supplement B is added around the matrix dome. Organoids are maintained under standard cell culture conditions for establishment and expansion.

Workflow Stage Key Process
Tissue Preparation Primary lung cancer tissue washing and fragmentation
Cell Isolation Enzymatic dissociation and filtration
3D Matrix Culture Cell embedding and extracellular matrix-supported growth
Organoid Expansion Culture maintenance using complete lung cancer organoid medium

Applications

XR Human Lung Cancer Organoid Culture Kit is designed for researchers developing patient-derived lung cancer organoid models and performing advanced 3D tumor research. Compared with traditional two-dimensional cancer cell culture models, lung cancer organoids provide a more physiologically relevant platform for studying tumor characteristics, treatment responses, and disease mechanisms.

Patient-Derived Lung Cancer Organoid Establishment

The kit supports the generation of lung cancer organoids from primary tumor tissues, providing researchers with a practical approach for establishing patient-derived tumor models for experimental studies.

  • Lung cancer organoid culture — Establish and maintain three-dimensional lung tumor organoid models from tumor-derived samples.

  • Patient-derived cancer model development — Generate experimental models that better represent individual tumor characteristics compared with conventional cell lines.
