Description

Multiplex probe-based qPCR laboratories often face two challenges at the same time. First, the detection of low-copy or single-copy targets demands very high sensitivity from the reagent. Second, in repeated multiplex runs, residual amplification products can contaminate new reactions and cause false positives — and in high sensitivity multiplex detection, contamination is exactly what can mask the real signal of the target.

StarLighter U+ HS Probe Multiplex qPCR Mix is a 2× premix that contains all components except the template, primers and probes. It combines a high-performance hot-start Taq Pro DNA polymerase with a buffer reaction system optimized for multiplex reactions, effectively inhibiting non-specific amplification and greatly increasing qPCR amplification efficiency and the sensitivity of multiplex detection. The UDG version (catalog No. FS-Q8003) introduces a dUTP/UDG anti-contamination system that works at room temperature, eliminating the impact of amplification-product contamination on qPCR results and ensuring accuracy. This high sensitivity multiplex probe qPCR master mix reaches a detection limit as low as a single copy.

To set up a reaction, simply add primers, template, probes, ROX (according to the instrument used) and water — the ROX reference dye is supplied with the kit. The kit provides good amplification curves across a wide concentration range, is suitable for quantification, qualitative detection and SNP detection of multiple target genes, offers good repeatability and high reliability, and is compatible with various types of qPCR probes.

Specification

Catalog No. Size
FS-Q8003-S 1 ml
FS-Q8003-01 5 ml

Format: 2× premix containing all components except template, primers and probes. The ROX reference dye is supplied with the kit and added according to the instrument used. The UDG version (FS-Q8003) contains a dUTP/UDG anti-contamination system that is active at room temperature.

Features

  • Detection limit as low as a single copy — high sensitivity multiplex probe qPCR master mix for rare and low-abundance targets.
  • Room-temperature dUTP/UDG anti-contamination system — eliminates the impact of amplification-product contamination on qPCR and ensures accurate results; UDG removal efficiency is higher than 99.9% (a difference of 10 Ct values corresponds to 99.9% removal efficiency), verified by quantifying the same dUTP-containing amplicon template with the UDG version (FS-Q8003) and a non-UDG version of the mix.
  • Hot-start Taq Pro DNA polymerase — effectively inhibits non-specific amplification and greatly increases qPCR amplification efficiency and multiplex detection sensitivity.
  • Multiplex performance with an internal control — in quadruplex detection of IPC (internal control, high template) together with hPRT1 (low GC), CSTB (high GC) and F8 (low GC) at 0.1 ng input each (about 30 copies), this master mix shows lower Ct values and higher fluorescence signals than a competitor product.
  • Low-concentration pathogen target detection — in quadruplex detection of IPC and African swine fever virus (ASFV) at about 20 copies, this master mix shows lower Ct values and higher fluorescence signals than a competitor product.
  • Wide linear range and higher detection sensitivity — in quadruplex detection of hPRT1, ApoE, actin and F8 with template inputs from 200 ng down to 0.0061 ng (six points), this master mix shows higher detection sensitivity and fluorescence signal intensity, with lower Ct values, than another competitor multiplex probe qPCR mix.
  • Good batch-to-batch consistency — three batches show consistent fluorescence intensity and Ct values in quadruplex quantitative amplification (10 ng, 1 ng and 0.1 ng inputs).
  • Broad primer/probe compatibility — primers and probes with different GC% and Tm values amplify well, compatible with various types of qPCR probes.
  • Good amplification curves across a wide concentration range — supports accurate quantification of multiple target genes with good repeatability and high reliability.
  • Simple 2× premix format — just add primers, template, probes, ROX (according to the instrument) and water.

Application

StarLighter U+ HS Probe Multiplex qPCR Mix (UDG) is suitable for:

  • Gene expression analysis — quantification of multiple target genes in a single multiplex reaction.
  • Low copy gene detection — sensitive detection of rare transcripts and low-abundance targets.
  • SNP and allele typing — qualitative and genotyping assays on multiple targets.

FAQ

What is StarLighter U+ HS Probe Multiplex qPCR Mix (UDG)?

It is a high sensitivity multiplex probe qPCR master mix in a 2× premix format. The UDG version (catalog No. FS-Q8003) combines a high-performance hot-start Taq Pro DNA polymerase and a multiplex-optimized buffer with a dUTP/UDG anti-contamination system that works at room temperature, for multiplex probe-based qPCR where high sensitivity and protection from amplification-product contamination are both required.

How does the dUTP/UDG anti-contamination system work?

The UDG version introduces a dUTP/UDG anti-contamination system that is effective at room temperature. It removes dUTP-containing amplification products that would otherwise contaminate new reactions, eliminating the impact of amplification-product contamination on qPCR results.

What is the difference between the UDG version (FS-Q8003) and a non-UDG version of this mix?

FS-Q8003-S and FS-Q8003-01 include the dUTP/UDG anti-contamination system. The UDG removal efficiency of the FS-Q8003 version is higher than 99.9% (a difference of 10 Ct values corresponds to 99.9% removal efficiency), verified by quantifying the same dUTP-containing amplicon template with the UDG version and a non-UDG version of the mix. Choose the UDG version when repeated multiplex testing makes protection from amplification-product contamination a priority. A non-UDG version of this mix (HS Probe Multiplex qPCR Mix, Universal) is also available in the StarLighter product line.

How sensitive is this multiplex qPCR master mix?

The detection limit can be as low as a single copy. In quadruplex detection, targets at 0.1 ng input (about 30 copies) and African swine fever virus (ASFV) at about 20 copies are detected with lower Ct values and higher fluorescence signals than competitor products.

Which polymerase is used, and how does it support multiplexing?

It uses a high-performance hot-start Taq Pro DNA polymerase combined with a buffer reaction system optimized for multiplex reactions. This configuration effectively inhibits non-specific amplification and greatly increases qPCR amplification efficiency and the sensitivity of multiplex detection.

Is the ROX reference dye included?

The ROX reference dye is supplied with the kit. During reaction setup, add ROX according to the instrument used, together with primers, template, probes and water.

How many targets can be detected, and can an internal control be included?

The master mix supports multiplex probe-based qPCR with multiple targets per reaction. Verified configurations include quadruplex detection of hPRT1, ApoE, actin and F8, as well as quadruplex detection of IPC (internal control) with hPRT1, CSTB and F8 in one reaction.

What template concentration range does this master mix cover?

Good amplification curves are obtained across a wide concentration range. In verified quadruplex runs, template inputs from 200 ng down to 0.0061 ng (six points) show higher detection sensitivity than a competitor multiplex probe qPCR mix, with good linearity across the gradient.

What applications is this master mix suitable for?

It is suitable for gene expression analysis, low copy gene detection, and SNP and allele typing, as well as quantitative and qualitative detection of multiple target genes with good repeatability and high reliability.

How does this master mix compare with the other StarLighter probe qPCR master mixes?

Within the StarLighter qPCR product line, this UDG version combines high sensitivity, multiplex capability and a dUTP/UDG anti-contamination system in one mix. For single-target probe assays, StarLighter offers the Probe qPCR Master Mix (Universal) and the High Sensitivity Probe qPCR Master Mix (Universal). For multiplex assays without a UDG requirement, the Probe-Based Multiplex qPCR Master Mix (Universal) is available; a non-UDG version of this mix (HS Probe Multiplex qPCR Mix, Universal) is also available under a separate catalog number.

What are the available catalog numbers and sizes?

The UDG version is available as FS-Q8003-S (1 ml) and FS-Q8003-01 (5 ml).
