Description

RIPA Lysis Buffer-high is a strong RIPA protein extraction buffer designed for efficient cell and tissue lysis when conventional extraction methods cannot fully solubilize target proteins. During protein preparation, incomplete cell disruption, poor membrane protein recovery, or low extraction efficiency from complex samples can affect downstream analysis. This high-strength RIPA lysis buffer provides enhanced protein solubilization capability to help researchers obtain reliable protein lysates for Western blot, SDS-PAGE, immunoprecipitation (IP), and co-immunoprecipitation (Co-IP) workflows.

Compared with mild or moderate lysis buffers, RIPA Lysis Buffer-high offers stronger disruption of cellular structures and improved extraction of difficult-to-solubilize proteins, including membrane-associated proteins and proteins tightly associated with cellular components. The resulting whole-cell lysates or tissue protein extracts are suitable for common molecular biology and biochemical research applications.

RIPA Lysis Buffer-high is part of a RIPA lysis buffer series with different extraction strengths, allowing researchers to select an appropriate protein extraction reagent according to sample type, protein characteristics, and experimental requirements.

Applications

RIPA Lysis Buffer-high is widely used as a protein extraction reagent in molecular biology, cell biology, and biochemical research workflows.

  • Western Blot Protein Analysis: Preparation of high-quality protein lysates for SDS-PAGE separation and Western blot detection of target proteins.

  • Cell Protein Extraction: Efficient lysis of adherent cells and suspension cells for whole-cell protein analysis.

  • Tissue Protein Extraction: Extraction of proteins from animal tissues and complex biological samples where stronger lysis is required.

  • Membrane Protein Extraction: Improved solubilization of membrane-associated proteins for downstream characterization.

  • Immunoprecipitation (IP): Preparation of protein lysates for antibody-based protein enrichment workflows.

  • Co-Immunoprecipitation (Co-IP): Supports studies involving protein-protein interactions and protein complexes.

  • SDS-PAGE Sample Preparation: Provides protein samples suitable for electrophoretic separation and analysis.

  • Biochemical Research: Used in protein expression studies, signaling pathway analysis, and cellular protein characterization.

Protocol Overview

For optimal protein extraction performance, samples should be processed under cold conditions.

  • Add appropriate protease inhibitors or phosphatase inhibitors before use according to experimental requirements.
  • Lyse samples on ice or at 4°C to minimize protein degradation.
  • Incubate samples with RIPA Lysis Buffer-high until complete cell disruption is achieved.
  • Centrifuge lysates at 10,000–14,000 ×g for 5 minutes and collect the clarified supernatant for downstream applications.

Because RIPA buffer contains detergents, protein concentration determination should be performed using detergent-compatible methods such as BCA or Lowry assays instead of Bradford assays.

FAQ

What is RIPA Lysis Buffer-high used for?

RIPA Lysis Buffer-high is a strong cell lysis buffer and protein extraction reagent used to extract proteins from cells and tissues for applications including Western blot, SDS-PAGE, immunoprecipitation, and Co-IP. It is especially useful when stronger protein solubilization is required.

What is the difference between strong, medium, and weak RIPA buffer?

The main difference is extraction strength. Strong RIPA buffer provides higher disruption capability for difficult-to-solubilize proteins, while weaker RIPA formulations are typically selected when preserving protein complexes or maintaining gentler extraction conditions is preferred.

Can RIPA Lysis Buffer-high be used for Western blot?

Yes. Protein lysates prepared with RIPA Lysis Buffer-high are commonly used for SDS-PAGE and Western blot analysis. The buffer enables efficient extraction of cellular proteins before electrophoresis and antibody detection.

Is RIPA buffer suitable for membrane protein extraction?

Strong RIPA lysis buffer can improve extraction of membrane-associated proteins because of its enhanced protein solubilization capability. Optimization may be required depending on the membrane protein characteristics.

Which protein assay method is recommended after RIPA extraction?

Because RIPA buffer contains detergents, Bradford assays may produce inaccurate results. BCA or Lowry protein assays are recommended for protein concentration measurement after RIPA extraction.

Why does RIPA lysate sometimes contain a transparent gel-like material?

A transparent gel-like precipitate may appear after RIPA lysis because of genomic DNA and DNA-protein complexes. This is a common phenomenon and usually does not affect downstream protein analysis after centrifugation and collection of the supernatant.
