SDS Lysis Buffer is a strong denaturing protein extraction buffer designed for efficient cell and tissue lysis when mild lysis reagents cannot fully release target proteins. During protein sample preparation, incomplete disruption of cellular structures, poor solubilization of membrane-associated proteins, or insufficient extraction efficiency from complex biological samples can affect downstream protein analysis. This SDS-based cell lysis buffer provides powerful protein solubilization capability to generate reliable protein lysates for Western blot, SDS-PAGE, and other protein analysis workflows.

Formulated with SDS detergent, Tris buffer, EDTA, and protein protection components, SDS Lysis Buffer enables rapid disruption of cellular membranes and efficient extraction of total proteins from animal cells, plant samples, bacteria, yeast, and tissue specimens. Compared with mild non-denaturing lysis buffers, this strong protein extraction buffer provides enhanced recovery of difficult-to-solubilize proteins and is particularly suitable for applications requiring complete protein denaturation and maximum protein yield.

SDS Lysis Buffer is optimized for preparing protein lysates for electrophoretic and antibody-based detection workflows. Due to the presence of SDS detergent, lysates prepared with this buffer are not suitable for Bradford protein assays and should be quantified using detergent-compatible protein determination methods when required.

Protocol Overview

For optimal protein extraction performance, samples should be processed under cold conditions to minimize protein degradation.

  • Add PMSF or other required protease/phosphatase inhibitors before use according to experimental requirements.
  • Lyse samples on ice or at 4 °C to maintain protein stability.
  • Add SDS Lysis Buffer to cells or tissues and ensure complete sample disruption.
  • Centrifuge lysates at 10,000–14,000 ×g for 3–5 minutes and collect the clarified supernatant for downstream analysis.

Because SDS-containing lysates contain high detergent concentrations, Bradford protein assays are not recommended. Detergent-compatible protein quantification methods should be selected when protein concentration measurement is required.
