Description

Laboratories measuring RNA across a broad concentration range need an assay that selectively detects RNA while accommodating different sample volumes and common laboratory contaminants. The StarLighter RNA BR Assay Kit provides a fluorescence-based method for broad range RNA quantification. It is highly selective for RNA over double-stranded DNA (dsDNA) and detects initial RNA concentrations from 1 ng/µL to 100 ng/µL, depending on sample volume, with a total assay range of 20–1000 ng.

This RNA Broad Range Assay Kit accepts 1–20 µL of sample in a final assay volume of 200 µL. The protocol can be performed with either a Qubit fluorometer or a fluorescence microplate reader. For microplate-based RNA quantification, fluorescence is measured at 620 nm excitation and 660 nm emission.

The assay tolerates common contaminants, including salts, free nucleotides, solvents, detergents, proteins, agarose, and PEG at the concentrations listed below. Signal development requires a 2-minute incubation at room temperature while protected from light. When protected from light, the fluorescent signal remains stable for up to 3 hours after incubation.

FAQ

What is the quantification range of the StarLighter RNA BR Assay Kit?

The assay detects initial RNA concentrations from 1 ng/µL to 100 ng/µL, depending on the sample volume. Its total assay range is 20–1000 ng.

How much RNA sample is required for each assay?

Each assay accepts 1–20 µL of RNA sample. The working solution volume must be adjusted so that the final assay volume is exactly 200 µL.

Can this RNA Broad Range Assay Kit be used with a Qubit fluorometer?

Yes. The protocol is compatible with a Qubit fluorometer. The assay requires two standards, QS0 and QS1, and thin-walled, clear 0.5 mL PCR tubes. On a Qubit 4 Fluorometer, RNA Broad Range is selected as the assay type.

Can the kit be used with a fluorescence microplate reader?

Yes. The assay supports a 96-well fluorescence microplate reader workflow. Use a black-wall, clear-bottom microplate to minimize cross-talk and set the reader to 620 nm excitation and 660 nm emission.

How long should the RNA assay incubate before measurement?

Incubate the prepared tubes or microplate for 2 minutes at room temperature, 20–25°C, while protected from light. The fluorescent signal remains stable for up to 3 hours after incubation when protected from light.

Is the assay selective for RNA over DNA?

Yes. The StarLighter RNA BR Assay is highly selective for RNA over double-stranded DNA.

Which StarLighter RNA BR Assay Kit sizes are available?

Catalog number FS-T1007-S contains materials for 100 reactions. Catalog number FS-T1007 contains materials for 1000 reactions.

How should the StarLighter RNA BR Assay Kit be stored?

The kit ships on ice. RNA BR Standards QS0 and QS1 must be stored at -25 to -15°C. All other kit components should be stored at 2–8°C. The fluorescent reagent, working solution, and assay tubes must be protected from light to minimize photobleaching.

When can I expect my order to ship?

Most orders are filled and shipped within 2-3 business days from the time they are received.

Our standard shipping usually take 2-5 days.

We also provide express shippping for time-sensitive deliveries.
