Product Description

The SHENTEK® REO-3 Analysis Kit is a qualitative assay serving as an analytical quality control reagent engineered exclusively for the molecular evaluation of bioprocessing inputs, such as in vitro cell culture supplements and cell banks. Optimized for compatibility with the SHENTEK® Nucleic Acid Extraction Kit (Product No. 1506730), this assay ensures the efficient recovery and analysis of target nucleic acids from complex industrial matrices.

Technical Specifications

Parameter Details
Intended use Qualitative detection of Reovirus 3 (REO-3) contamination in cell banks, virus seed banks and bovine serum
Assay sensitivity / Limit of detection (LOD) 50 copies per reaction
Specificity No cross-amplification with bovine, porcine genomes, common production/engineering cell lines (CHO, Vero, 293T, MDCK, NS0, Sf9) or engineering bacteria (E. coli, Pichia pastoris)
Reaction volume 20 μL total reaction volume (10 μL RT-qPCR MIX + 10 μL sample or control)
Thermal cycling conditions Reverse transcription: 50 °C 15:00 (1 cycle); Activation: 95 °C 00:30 (1 cycle); Denaturation: 95 °C 00:15; Annealing/extension: 60 °C 01:00; 45 cycles (fluorescence read during annealing/extension)
Controls and interpretation thresholds Controls: NTC, NCS, PCS, Internal Positive Control (IPC). Typical Ct acceptance criteria: FAM (target): Positive if Ct < 40.00 (at least one well) or Ct < 40 with duplicate amplification; VIC (IPC): Ct < 35.00 indicates valid IPC amplification. NTC/NCS: no specific amplification (Ct ≥ 40 or absent) required. See user guide tables for full interpretation rules.
Kit size / throughput Reagents for 50 reactions
Storage & stability Store components at -20 °C (recommended). Kit components stable for up to 24 months; items with recommended storage at -20 °C should be maintained within ±10 °C (–30 °C to –10 °C).
Compatible instruments SHENTEK-96S, LightCycler 480 II, ABI 7500, Bio-Rad CFX96 and other real-time PCR systems supporting probe assays and user-defined cycling

Features

  • High sensitivity: Analytical sensitivity down to 50 copies per reaction (LOD).
  • High specificity: No cross-amplification with common cell lines (CHO, Vero, 293T, MDCK, NS0, Sf9) or bacterial/yeast genomes (E. coli, Pichia pastoris), and with bovine/porcine genomes.
  • Complete RT-qPCR solution: Includes primer & probe mix, RT-qPCR enzyme mix, buffer and internal positive control mix for end-to-end detection when combined with recommended extraction kit.
  • Compatible with multiple instruments: Validated on several common real-time PCR platforms and supports standard probe-based RT-qPCR setup.
  • Designed for complex matrices: Optimized to detect REO-3 RNA from cells and bovine serum when used with SHENTEK® Virus DNA & RNA Extraction Kit.
  • Internal control included: IPC MIX provided to monitor extraction and PCR inhibition.

Applications

  • Qualitative molecular analysis of REO-3 target sequences in industrial cell banks
  • Analytical quality control of biological seed lots
  • Molecular evaluation of in vitro cell culture supplements for REO-3 nucleic acids
  • In-process monitoring and lot-release analysis for biological manufacturing
  • In vitro research projects assessing REO-3 target sequences in complex culture matrices

Attention

• For Research Use Only (RUO). Not for use in diagnostic procedures.

• Read Material Safety Data Sheets (MSDS) and wear appropriate PPE (eyewear, mask, gloves, protective clothing).

• Handle positive control and extraction in a biosafety cabinet and follow BSL-2 safety guidelines.

• Use nuclease-free consumables and certified RNase/DNase-free reagents and tips to avoid contamination.

• Thaw kit completely at 2–8 °C or on ice, vortex and centrifuge briefly before use.

• Protect primer & probe mixes from light.

• Store components at -20 °C; short-term storage below -30 °C is acceptable if supported by stability data.

• Check expiration dates on labels prior to use.

• If IPC (VIC) signal indicates inhibition, repeat sample preparation or remove inhibitors and repeat assay.

Frequently Asked Questions (FAQ)

Q1: What is the assay's limit of detection (LOD)?
The assay sensitivity is 50 copies per reaction.
Q2: Which sample types are supported?
Validated sample types include cell culture (up to 1×10^7 cells per 100–200 μL) and bovine serum. For other matrices consult technical support and use the recommended SHENTEK® extraction kit.
Q3: What controls are required and how are results interpreted?
Run NTC (No Template Control), NCS (Negative Control Sample), PCS (Positive Control Sample) and IPC. Typical acceptance thresholds: FAM target Ct < 40 indicates positive amplification; IPC (VIC) Ct < 35 indicates successful internal control amplification. Follow the user guide control tables for detailed interpretation.
Q4: Which instruments are compatible?
Compatible with common real-time PCR systems including SHENTEK-96S, LightCycler 480 II, ABI 7500, Bio-Rad CFX96 and other instruments that support probe-based RT-qPCR.
