Description

As a master regulator of Th2 immunity, IL-4 activates the JAK1/3-STAT6 signaling cascade via the IL-4Rα/γc receptor complex, directing naive CD4+ T-cell differentiation into Th2 effectors secreting IL-5/IL-13 while mediating B-cell class switching to IgE/IgG1 and FcεRI upregulation.

The recombinant human IL-4-His fusion protein is a high-purity functional cytokine produced in HEK-293 expression systems, featuring a C-terminal hexahistidine tag to facilitate affinity purification and structural integrity.

In cell therapy applications, this protein serves as an essential inducer for monocyte-derived immature dendritic cell (imDC) generation through GM-CSF synergy, effectively suppressing CD14 expression and enhancing antigen-capturing capacity to standardize tumor vaccine production.

It also modulates CAR-T cell manufacturing by balancing Th1/Th2 polarization for optimized solid tumor responses and prolongs mast cell survival in vitro to enhance stability of allergen-specific immunotherapies.

The HEK-293-derived expression ensures authentic glycosylation patterns, while the His-tag maintains native conformation and enables high-resolution chromatographic purification.

Features

  • High Biological Potency (TF-1 Validated)

Demonstrates strong proliferative activity in TF-1 cell assays (7.9 × 106 IU/mg), ensuring reliable Th2-skewing and DC induction performance.

  • HEK293 Expression with Authentic Glycosylation

Mammalian expression preserves native folding and glycosylation, supporting consistent IL-4Rα/γc signaling activation.

  • Low Endotoxin, Cell-Therapy Compatible

Endotoxin level < 0.1 EU/µg and mycoplasma-negative, suitable for immune cell culture and CGT research workflows.

  • Batch-to-Batch Consistency

Manufactured under controlled QC standards with verified purity (>90% SDS-PAGE), minimizing experimental variability.

Applications

Monocyte-Derived Dendritic Cell (imDC) Generation

Used synergistically with GM-CSF to suppress CD14 expression and drive standardized immature DC differentiation.

T-Cell Polarization Studies

Directs naïve CD4⁺ T cells toward Th2 lineage via STAT6 activation, supporting immune-profiling research.

CAR-T / CGT Process Development

Modulates Th1/Th2 balance during T-cell manufacturing to optimize functional phenotype for solid tumor research.

Mast Cell & Allergy Research Models

Prolongs mast cell survival in vitro, enabling stable allergen-response and IgE-related studies.

Stability & Storage

  • Lyophilized powder: Store at −20 °C in the original sealed container.
  • Reconstitution: Reconstitute with sterile Water for Injection, 0.9% NaCl, or PBS (pH 7.4) prior to use.
  • Handling: Aliquot after reconstitution and avoid repeated freeze–thaw cycles.

Cautions

• Research Use Only: This product is not intended for diagnostic or therapeutic use.

• Handling: Prepare solutions using sterile technique. Avoid microbial contamination to maintain activity.

• Storage: Follow recommended storage conditions. Repeated freeze–thaw cycles may reduce performance.
