Rat Primary Cardiomyocytes from Cell Biologics are derived from the heart of pathogen-free laboratory post neonatal mice (day 1-3).  Cells are freshly isolated and shipped at passage 1. Cells are grown in T25 tissue culture flasks, 6-well plates or 12-well plates, which were pre-coated with gelatin-based solution and fibronectin and incubated in Cell Biologics’ Culture Complete Growth Medium for 3-7 days. The beating cardiomyocytes were shipped and ready for customers’ experiments. The method we use to isolate primary cardiomyocytes cells was developed based on a combination of established and our proprietary methods. These cells are purified by removing fibroblasts and endothelial cells. These cells will not proliferate in culture and cannot be passaged.

Rat Primary Cardiomyocytes from Cell Biologics display typical elongated and flattened morphology under light microscopy. Cells are tested for expression of cardiomyocyte marker using antibody cardiac Troponin I (cTnI, Catalog No. ab47003, from Abcam) or Myosin heavy chain (Catalog No. MA5-35613, from Thermofisher. Scientific) by immunofluorescence staining. All cells test negative for mycoplasma, bacteria, yeast, and fungi. HIV-1, hepatitis B and hepatitis C are not detected for all donors and/or cell lots. Per request, a Certificate of Analysis will be provided for each cell lot purchased.

Rat Primary Cardiomyocytes can be used for standard biochemical procedures including RT-PCR, Western blotting, immunoprecipitation, immunofluorescent staining, immunofluorescent flow cytometry, or generating cell derivatives for desired research applications.

Storage

Cell Biologics ships with fresh complete culture medium. On receipt, cells are ready for experimental use. Cells can be maintained up to 7 days under the cell culture conditions specified by Cell Biologics. Subculturing, freezing and thawing of cardiomyocytes are not recommended.
