Descprition

This product uses a unique lysis buffer system to rapidly release RNA from cultured cell samples for RT-qPCR reactions, eliminating the time-consuming and laborious RNA purification, and only takes 7 minutes to obtain the required RNA template. Used with the 5× Direct RT Mix and 2× Direct qPCR Mix-SYBR provided in the kit, effective real-time quantitative PCR results can be quickly obtained.

5× Direct RT Mix and 2× Direct qPCR Mix-SYBR have strong inhibitor tolerance, enabling efficient reverse transcription and specific amplification using the sample lysate as template. The kit contains high-RNA-affinity Foregene Reverse Transcriptase, as well as Hot-Start T-Taq DNA Polymerase, dNTPs, MgCl₂, reaction buffer, PCR optimizer and stabilizer. When used with the lysis buffer, they enable rapid and simple sample detection with high sensitivity, strong specificity, and excellent stability.

The kit is designed for micro-scale lysis of cells cultured in 96-well plates, offering excellent uniformity and consistency. The kit components provide 96 lysis reactions, 96 reverse transcription reactions, and 96×2 qPCR reactions, supporting one-batch processing of an entire 96-well cell plate. Avoid contamination and performance degradation of reagents caused by repeated opening, freeze-thaw cycles.

Components

• Buffer CL: Provides optimal conditions for cell lysis reactions.

• Foregene Protease Plus II: Lyses cells and releases nucleic acids in the lysis buffer.

• DNA Eraser: DNA remover, to eliminate the impact of genomic DNA on subsequent experiments.

• Buffer ST: Terminates the active substance in lysate to avoid effects on subsequent RT.

• 5× Direct RT Mix: Contains high-RNA-affinity Foregene Reverse Transcriptase, RNase Inhibitor, dNTPs, stabilizers, enhancers, optimizers, and optimized reverse transcription primers (Random Primer, Oligo(dT) 18 Primer).

• 2× Direct qPCR Mix-SYBR: This reagent contains Foreasy HS Taq DNA Polymerase, dNTPs, MgCl₂, reaction buffer, SYBR Green I, PCR optimizer and stabilizer.

• 50× ROX Reference Dye: Generally used on real-time quantitative PCR instruments of ABI, Stratagene and other companies, it is used to adjust the difference between PCR tubes caused by PCR pipetting errors. The 50× ROX Reference Dye concentration required for different instruments is different, and the user can add it according to the recommended concentration of the instrument.

• RNase-Free ddH₂O: RNase-Free sterilized ultrapure water for two-step RT-qPCR reactions.

• Pay attention to the operation of the experiment to avoid cross-contamination between samples.

• Keep the experimental environment and utensils clean to avoid RNase contamination and RNA degradation.

• Please prepare the cell lysis system fresh and use it immediately.

• Avoid repeated freeze-thaw cycles for 2× Direct qPCR Mix-SYBR, as this may reduce PCR efficiency.
