Principle of the Assay

Pandy’s Reagent contains phenol dissolved in water. Upon contact with CSF, phenol induces precipitation of proteins—primarily globulins, but also albumin and other protein fractions. The resulting degree of turbidity correlates with protein concentration and is visually classified into graded categories ranging from negative to strongly positive reactions.

Specificity & Limitations

Pandy’s reaction is not globulin-specific and may also respond to albumin and other protein fractions present at elevated concentrations. For enhanced specificity toward globulins, it is recommended to perform an additional confirmatory assay, such as the Nonne–Apelt reaction.
