Oil Red O

Procedure 

  • Prepare a 0.2% (w/v) Oil Red O stock solution in methanol. 
  • Prepare staining solution by mixing 35 ml of stock solution from step 1 with 10 ml of 
    1N NaOH. Filter through coarse ashless filter paper immediately before use. 
  • After electrophoresis, soak cellulose acetate strip in staining solution for 1 1/2 hours. 
  • Pour off excess stain and gently wipe any precipitate off the cellulose acetate strip. 
  • Immerse the strip in destaining solution (1 part methanol and 3 parts glycerol) for 10 
    minutes. Remove excess destain, blot dry, spray with clear acrylic spray coating for 10 
    seconds and allow to dry.
