Target Background | A c-myc Tag is a polypeptide tag derived from the c-myc gene product that can be added to a protein using recombinant DNA technology. It can be used for affinity chromatography to separate recombinant, overexpressed protein from wild-type protein expressed by the host organism. Additionally, it aids in the isolation of protein complexes with multiple subunits. A c-myc Tag is widely used in assays requiring recognition by an antibody. If no antibody against the studied protein is available, adding a myc-tag allows tracking of the protein using an antibody against the Myc epitope. Examples include cellular localization studies by immunofluorescence or detection by Western blotting. The tag can be fused to the C-terminus or N-terminus of a protein, though it is advisable not to fuse it directly behind the signal peptide of a secretory protein, as this may interfere with translocation into the secretory pathway. |
