Synonym(s): 3-(N-Morpholino)propanesulfonic acid, 4-Morpholinepropanesulfonic acid, MOPS, Free Acid, ULTROL® Grade, 3-Morpholinopropane sulfonic acid

Description

In biological experiments, it is an important pH stabilizing reagent. Typically, a suitable weak acid is mixed with its conjugate base to obtain the appropriate pH value. Most biological reactions occur under neutral conditions, generally with a pH between 6 and 8, so the effective buffering range of the buffer is also required to be between 6 and 8. In addition, it is required that the acidic and basic forms of the buffer do not chelate with certain metal ions.

Features

  • High-purity biochemical grade (≥99%)

Ensures reliable performance for molecular biology, biochemistry, cell culture, and analytical applications requiring strict quality control.

  • Optimal buffering near physiological pH

With a pKa of approximately 7.2, MOPS provides excellent buffering capacity in the 6.5–7.9 range, ideal for biological reactions conducted under near-neutral conditions.

  • Low UV absorbance for sensitive assays

Exhibits minimal absorbance in the UV range, ensuring compatibility with spectrophotometric, nucleic acid, and protein quantification assays without introducing interference.

  • Minimal metal-ion binding

As a Good’s buffer, MOPS shows negligible chelation with divalent metal ions, maintaining the integrity of metal-dependent enzymes and minimizing experimental artifacts.

  • High stability and excellent solubility

Available as a stable, pure white powder that dissolves easily in water to produce clear, colorless solutions. Suitable for long-term storage at room temperature.

Applications

  • Electrophoresis buffers

Widely used in RNA and DNA gel electrophoresis (such as MOPS running buffer for formaldehyde agarose gels) to ensure stable pH and high-resolution band separation.

  • Cell culture media buffering

Suitable for maintaining physiological pH in mammalian, bacterial, or yeast culture media, particularly in CO₂-independent or serum-reduced systems.

  • Protein purification workflows

Used in chromatography buffers (ion-exchange, SEC, affinity purification) due to its stable buffering action and non-chelating properties that preserve protein structure and activity.

  • Enzyme reactions and biochemical assays

Ideal for enzymatic kinetics, activity measurements, and biochemical reactions requiring precise pH control near neutrality.

  • Sample preparation and extraction

Utilized in buffers for cell lysis, nucleic acid extraction, and protein isolation where consistent pH and low interference are essential for downstream analysis.

  • Metal-ion-sensitive experiments

Recommended for assays involving metal-dependent enzymes or pathways where avoiding metal ion chelation is critical for accurate results.

FAQ

What is MOPS buffer used for in molecular biology experiments?

MOPS buffer is a widely used Good’s buffer in molecular biology and biochemistry to maintain a stable pH environment between 6.5 and 7.9. It is commonly applied in RNA electrophoresis, enzyme assays, protein purification, and cell culture systems where physiological pH stability is required.

Why is MOPS preferred for RNA and protein experiments?

MOPS is preferred because it provides stable buffering near neutral pH, has low UV absorbance, and does not strongly interact with metal ions. These properties help maintain RNA integrity, protein stability, and consistent enzymatic activity during experimental workflows.

What is the difference between MOPS and other biological buffers like HEPES or Tris?

Compared with Tris and HEPES, MOPS offers a more defined buffering range around physiological pH and lower metal ion interaction. This makes it particularly suitable for RNA electrophoresis and enzyme-based assays requiring high reproducibility.

Can MOPS buffer be used in cell culture applications?

Yes. MOPS is commonly used in CO₂-independent and serum-reduced cell culture systems to maintain stable pH. It helps preserve consistent physiological conditions in mammalian, bacterial, and yeast cell experiments.

Does MOPS buffer interfere with enzyme activity or metal ions?

MOPS is a non-chelating buffer and shows minimal interaction with divalent metal ions such as Mg²⁺ and Ca²⁺. This makes it suitable for metal-dependent enzyme assays and biochemical reactions where ion availability must remain unchanged.

How should MOPS buffer be stored for long-term stability?

MOPS powder should be stored at room temperature in a dry, tightly sealed container. Proper storage prevents moisture absorption and ensures long-term chemical stability and consistent buffering performance.
