specifically designed for the in vitro induction of human mesenchymal stem cells (hMSCs) into chondrogenic. It can be used for the chondrogenic differentiation of hMSCs derived from bone marrow, umbilical cord, adipose tissue, and other sources.

Storage Conditions and Validity Period

  • Human Mesenchymal Stem Cell Chondrogenic Differentiation Basal Medium: Store at 2°C~8°C,valid for 12 months.
  • Human Mesenchymal Stem Cell Chondrogenic Differentiation Supplement: Store at -25°C~-10°C,valid for 12 months.
  • Prepared Human Mesenchymal Stem Cell Chondrogenic Differentiation Medium: Store at 2°C~8°C, valid for 2 weeks.

 

Precautions

  • Upon first thawing, aliquot it to avoid damage via repeated freeze-thaw cycle,and ensure sterile handling during use.
  • For research use only.
  • The chondrogenic differentiation level of human mesenchymal stem cells may vary due to differences in cell sources, donors, and pre-culture conditions.

 

Directions for Use

1.Thaw 5 mL of Human Mesenchymal Stem Cell Chondrogenic Differentiation Supplement, add it to 195 mL of Human Mesenchymal Stem Cell Chondrogenic Differentiation Basal Medium, and prepare 2.5% Human Mesenchymal Stem Cell Chondrogenic Differentiation Medium (adjust volumes according to requirements).

2.Resuspend recovered/digested hMSCs in Human Mesenchymal Stem Cell Chondrogenic Differentiation Medium at a concentration of 0.5×10 6 ~1.0×10 6 cells/mL. Transfer 1mL of cell suspension into a 15 mL polypropylene centrifuge tube, centrifuge at 500 ×g for 5 minutes to aggregate the cells, remove the supernatant, and add another 1 mL of Human Mesenchymal Stem Cell Chondrogenic Differentiation Medium.

3.Place the centrifuge tube in a cell culture incubator, loosen the tube cap for air exchange.

4.Spheroids will form within 24~48 hours, and the size of the spheroids is related to the seeded cell quantity.

5.Change the medium every 2~3 days, 1mL each time. Gently pipette the spheroids to avoid adherence to the tube bottom.

Note: Gently change the medium to avoid sucking out the spheroids.

6.After the third week of differentiation, sections could be stained to observe the results.
