Description

Laboratories working with fluorescent human kidney fibroblasts need clearly documented tissue origin, reporter generation, cell format, culture requirements, characterization, contamination testing, shipping, and storage information before planning an experiment. Immortalized RFP-Expressing Human Primary Kidney Fibroblasts provide a cryopreserved, human kidney tissue-derived fibroblast culture with red fluorescent protein expression.

The cells are isolated from human kidney tissue and grown in gelatin-precoated tissue culture flasks with Cell Biologics' Complete Growth Medium. RFP-expressing stable cells are generated by transduction with RFP lentiviral particles. Red fluorescent protein produces bright red fluorescence when exposed to yellow-green light.

Cells are harvested at passage 2-3 and cryopreserved in vials. Each vial contains 0.5 106 cells per mL and is delivered frozen. The immortalized human kidney fibroblasts display spindle morphology and are characterized by immunofluorescence staining with anti-fibronectin antibodies.

Under the cell culture conditions specified by Cell Biologics, the cells can be expanded for more than 10 passages using a split ratio of 1:2. Repeated freezing and thawing are not recommended.

Research Applications

These immortalized RFP-expressing human kidney fibroblasts are intended for laboratory research that requires human kidney tissue-derived fibroblasts with an intrinsic red fluorescent reporter. The documented product characteristics support the following research workflows without making a disease-specific, diagnostic, or therapeutic performance claim.

  • RFP fluorescence observation: Observation of RFP-expressing cells using yellow-green excitation appropriate for red fluorescent protein.
  • Fluorescent cell culture workflows: Research workflows requiring an RFP-expressing human kidney fibroblast culture.
  • Fibroblast characterization workflows: Evaluation of spindle morphology and anti-fibronectin immunofluorescence staining.
  • Extended cell expansion: Experiments requiring expansion beyond the limited passage range typical of primary cultures, using the specified culture conditions.
  • Human kidney fibroblast research: In vitro research requiring fibroblasts derived from human kidney tissue.

The supplied documentation does not state a disease-specific application, RFP-positive percentage, fluorescence stability by passage, or a product-specific immortalization method. Suitability must be determined by the investigator for the intended experimental design.

Culture Information

  • Use M2267 Complete Fibroblast Medium w/ Kit - 500 mL as the suggested medium.
  • Grow the cells in gelatin-precoated tissue culture flasks with complete growth medium.
  • Use a recommended split ratio of 1:2 under the specified cell culture conditions.
  • The cells can be expanded for more than 10 passages under those conditions.
  • Avoid repeated freezing and thawing.

Shipping, Storage, and Handling

  • Cryopreserved cells are shipped overnight on dry ice.
  • Upon arrival, transfer the frozen vial immediately to liquid nitrogen at approximately -180 C until ready for use.
  • Do not store the delivered frozen cells in a -20 C or -80 C freezer.
  • Live cell shipment is available on request.
  • Handle the cells with caution and treat primary cell-derived products as potential pathogens because no test procedure can completely guarantee the absence of infectious agents.
