Description

When extended cell culture and direct fluorescence observation are required, conventional primary cells may not provide the lifespan or existing fluorescent signal needed for the planned workflow. Immortalized RFP-Expressing Canine Primary Colonic Smooth Muscle Cells provide a primary-derived, extended-lifespan canine colonic smooth muscle cell model with red fluorescent protein expression.

These cells are isolated from the colonic tissue of beagle dogs and cultured in gelatin pre-coated tissue culture flasks with Cell Biologics' Complete Growth Medium. Cells at passages 2–3 are harvested and cryopreserved in vials. Each vial contains 0.5 × 106 cells per mL and is delivered frozen.

The canine colonic smooth muscle cells are characterized by immunofluorescence staining with an α-smooth muscle actin antibody. Testing is reported negative for bacteria, yeast, fungi, and mycoplasma.

RFP-expressing cells are developed through transduction with RFP-lentiviral particles. Red fluorescent protein produces bright red fluorescence when exposed to yellow-green light.

Under the cell culture conditions specified by Cell Biologics, these immortalized RFP-expressing canine colonic smooth muscle cells can be expanded for more than 10 passages at a split ratio of 1:2. Repeated freezing and thawing is not recommended.

Frequently Asked Questions

What are Immortalized RFP-Expressing Canine Primary Colonic Smooth Muscle Cells?

They are primary-derived immortalized smooth muscle cells isolated from beagle dog colonic tissue. The cells express RFP and are supplied as cryopreserved cells for research use only.

Are these primary cells or immortalized cells?

They are derived from canine primary colonic smooth muscle cells and subsequently immortalized to provide an extended lifespan. They should therefore be treated as an immortalized cell model derived from primary cells.

What species and tissue are the cells derived from?

The cells are isolated from the colonic tissue of beagle dogs.

How is RFP introduced into the cells?

According to the product datasheet, RFP-expressing cells are developed through transduction with RFP-lentiviral particles.

Which immortalization method is used for D-6001IM.RFP?

The manufacturer states that its immortalized cells are generated by overexpression of SV40 large T-antigen or hTERT. The supplied datasheet does not specify which method applies to catalog D-6001IM.RFP.

At what passage are the cells cryopreserved?

Cells at passages 2–3 are harvested and cryopreserved in vials.

What cell concentration is documented?

The datasheet states that each vial contains 0.5 × 106 cells per mL and is delivered frozen.

How are the canine colonic smooth muscle cells characterized?

The cells are characterized by immunofluorescence staining with an α-smooth muscle actin antibody.

What microbial quality control testing is documented?

The product datasheet reports that the cells are negative for bacteria, yeast, fungi, and mycoplasma.

How many passages can the cells be expanded?
