Description

Laboratories isolating human NK cells from whole blood need to remove non-NK cells while keeping the target NK cells unlabeled for downstream experiments. The K005 Human NK Cell Purification Cocktail provides a negative-selection workflow for human whole blood. Its antibodies recognize specific surface markers on non-target cells and crosslink these cells to multiple red blood cells, forming dense immunorosettes.

During density gradient centrifugation, the immunorosettes sediment together with free red blood cells. The target human NK cells remain unlabeled and become highly enriched at the interface between the plasma and the density gradient medium, where they can be collected for subsequent laboratory use.

This human NK cell purification cocktail is supplied as one kit in a 3 mL inner container measuring 16 mm × 35 mm. The product weight is 5 g. Before use, reconstitute the cocktail with 2 mL of sterile water. The recommended working amount is 40 µL of cocktail per 1 mL of human whole blood.

Important: Density gradient separation medium is not included with this product and must be supplied separately by the user.

What centrifugation conditions are used for density gradient separation?

After layering the diluted sample onto the density gradient medium, centrifuge at 1200 × g for 20 minutes using acceleration setting 1 and deceleration setting 0. Harvest the buffy coat layer and wash at 300 × g for 10 minutes using acceleration setting 8 and deceleration setting 6. Repeat the wash step once.

What blood volumes can be processed?

The workflow without preparation of autologous inactivated plasma specifies up to 15 mL of fresh whole blood per tube. The workflow that includes preparation of autologous inactivated plasma specifies up to 50 mL of fresh whole blood per tube.
