Description
Fluorescence-based primary cell studies require a clearly defined cell source, stable fluorescent labeling, documented characterization, and a reproducible culture workflow. These GFP-expressing mouse primary colonic smooth muscle cells provide a cryopreserved primary cell model derived from the colon tissue of pathogen-free laboratory C57BL/6 mice.
The cells are isolated from mouse colon tissue and cultured in gelatin-precoated tissue culture flasks using complete smooth muscle cell growth medium. Cells are harvested at passage 2–3 and cryopreserved for frozen delivery. Each vial contains 0.5 × 106 cells per mL.
These GFP-expressing primary cells are generated by transduction with GFP lentiviral particles. Green fluorescent protein produces bright green fluorescence when exposed to blue light, supporting experimental workflows that require fluorescent visualization of primary colonic smooth muscle cells.
Cell identity is characterized by immunofluorescence staining with an antibody against α-smooth muscle actin (α-SMA). Only the amount of complete culture medium required for immediate use should be prewarmed to 37°C. Repeated warming of complete medium is not recommended.
Culture Information
Culture Vessel Preparation
Coat sterile culture flasks or dishes with a gelatin-based coating solution for approximately 2 minutes. Aspirate the excess coating solution before seeding the cells.
Recovery from Cryopreservation
Thaw the cryovial rapidly in a 37°C water bath for less than 1 minute, or until only a small amount of ice remains. Transfer the cells into prewarmed culture medium and centrifuge at 200 × g for 5 minutes to remove residual cryopreservation medium. Resuspend the cell pellet and seed it into a gelatin-precoated T25 flask.
Incubation
Maintain the cells in a humidified incubator at 37°C with 5% CO2. Following initial seeding, leave the culture undisturbed for approximately 12–16 hours.
Medium Replacement
Replace the medium on the day following recovery and regularly thereafter to remove non-adherent cells and replenish nutrients. Cells should be examined daily for confluence and appropriate morphology.
Subculture
For expansion, wash the adherent cells with calcium- and magnesium-free phosphate-buffered saline and detach them using prewarmed 0.25% Trypsin-EDTA. The recommended split ratio for primary cells is 1:2.
Storage
Cryopreserved cells are shipped frozen on dry ice. Immediately upon arrival, transfer the vial to a liquid nitrogen storage tank until the cells are ready for use.
Do not use a standard −20°C or −80°C freezer for long-term storage of the cryopreserved primary cells. Live-cell shipment may also be available on request.
