General cell culture:
Supports the growth and maintenance of a variety of human and animal cell lines, including primary cells, fibroblasts, and tumor cell cultures.

Tissue and organ culture:
Due to its inclusion of nucleosides and coenzymes (such as NAD, NADP, and Coenzyme A), CMRL-1066 is suitable for short-term maintenance of mammalian tissue fragments or organ explants in vitro.

Low-serum or serum-free systems:
The presence of ascorbic acid, glutathione, cholesterol, and Tween 80 helps maintain membrane stability and cellular vitality under low-serum conditions. Hence, it is frequently used in the development of serum-free or chemically defined culture systems.

Host–pathogen co-culture studies:
As a nutritionally rich basal medium, CMRL-1066 is often employed for the co-culture of cell-dependent microorganisms such as Mycoplasma, Rickettsia, or certain viruses.
