Bf-001AGBM Sheep Anti-Rat Isolated GBM Serum

Description

Establishing an experimental anti-GBM nephritis model requires an antiserum that targets the rat glomerular basement membrane and provides sufficient material for a defined number of animals. Biofargo Bf-001AGBM is a sheep anti-rat isolated glomerular basement membrane serum supplied for the induction of passive anti-GBM nephritis in rats.

This nephrotoxic anti-GBM serum is supplied as 25 mL in 0.02 M phosphate-buffered saline at pH 7.3. One bottle contains sufficient antibody to induce passive anti-GBM nephritis in at least 25 rats weighing 175–200 g when used according to the stated directions.

The rat anti-GBM nephritis model is associated with mesangial proliferation, inflammatory cell vasculitis, glomerulosclerosis and renal fibrosis. The disease response is dose-dependent, so the complete dose must be delivered. Because antiserum lots, rat sources and experimental requirements can vary, a dose-response study should be performed to establish the desired disease severity for each lot.

Bf-001AGBM replaces the previous sheep anti-whole-rat-glomeruli serum. Both products target rat GBM and produce the same type of disease model. The current product has not been validated or recommended for use in mice.

Model Characteristics Described in the Product Data

Following administration of the anti-GBM serum, linear deposits of heterologous sheep IgG can become detectable in the glomerular basement membrane by immunofluorescence within minutes and increase in intensity during the following 3–5 days. Rat C3 can also localize in a linear pattern, and proteinuria can develop after 24 hours.

The autologous phase can become noticeable 8–10 days after anti-GBM antibody administration, with increased proteinuria and linear glomerular localization of rat IgG. By three weeks, the product data state that proteinuria can exceed 200 mg per 24 hours and that glomerular lesions can become apparent and increase in severity over time, leading to glomerulosclerosis.
