Product Description

The SHENTEK® MVM Detection Kit is a qualitative real-time PCR assay for detection of Minute Virus of Mice (MVM) contamination, covering MVM strains i, p, m and c, in mouse-derived cell lines and related sample matrices used in biopharmaceutical production (examples: CHO, BHK, NS0). The kit is designed for use with the SHENTEK® Virus DNA & RNA Extraction Kit (Product No. 1506730). The assay includes MVM-specific primer/probe mix, internal process control (VIR Internal Control), and positive control to monitor extraction and amplification. Combined with SHENTEK extraction, the kit enables efficient recovery and sensitive detection of MVM DNA from complex matrices such as cells and serum. The assay demonstrates high specificity (no cross-amplification observed with tested cell lines and microbial genomes) and a reported analytical sensitivity of 50 copies per reaction. The kit is supplied for 50 reactions.

Acceptance criteria — control wells NTC: FAM undetected; VIC: Ct < 35 for duplicate runs. NCS: FAM undetected; VIC: Ct < 35. PCS: FAM Ct < 35 and VIC Ct < 35
Acceptance criteria — test samples Positive: FAM Ct < 40 (at least one well) with specific amplification and VIC Ct < 35 in duplicates. Negative: FAM undetected in duplicates and Ctsample - CtNCS < 2. Not conclusive / possible inhibition: Ctsample - CtNCS ≥ 2 (compare VIC Ct of sample to NCS); retest or remove inhibitors
Compatible instruments (examples) SHENTEK-96S, LightCycler 480 II, ABI 7500 and most real-time PCR systems supporting probe assays
Storage & stability Kit components stored under recommended conditions (primarily -20 °C where indicated). Components stable up to 24 months (check label expiration date)

Features

  • Broad MVM strain coverage: Targets and detects four MVM strains (i, p, m and c).
  • High analytical sensitivity: Reported limit of detection of 50 copies per reaction.
  • High specificity: No cross-amplification with common mammalian cell lines or tested microbial genomes.
  • Internal process control included: VIR Internal Control (IC) monitors extraction and PCR inhibition.
  • Supplied controls: MVM Positive Control provided (reference product number supplied separately); NTC and NCS workflows specified.
  • Compatible with automated extraction: Recommended for use with rHCDpurify System and the SHENTEK® Virus DNA & RNA Extraction Kit to efficiently recover viral DNA from complex matrices.
  • Ready-to-use primer/probe mix: Pre-mixed MVM primer & probe simplifies setup and reduces assay variability.
  • Validated qPCR program: Predefined cycling conditions and instrument setup guidance provided for common platforms.

Applications

  • Contamination screening of mouse-derived cell lines (e.g., CHO, BHK, NS0) used in biopharmaceutical production
  • Testing of cell culture supernatant and serum for MVM DNA
  • In-process and release testing support in research environments
  • Research studies requiring qualitative detection of MVM

Attention

• For Research Use Only — consult compliance statement.

• Read Material Safety Data Sheets (MSDS) and follow handling instructions.

• Wear appropriate PPE (protective eyewear, mask, clothing and gloves).

• Handle positive control and extraction in a biological safety cabinet and follow BSL-2 laboratory safety guidelines.

• Use nuclease-free consumables and certified reagents.

• Thaw kit components at 2–8 °C or on ice; vortex and briefly centrifuge before use.

• Avoid cross-contamination: use separate areas and aerosol-resistant tips for reagent preparation, sample handling, and amplification.

• If VIC internal control Ct of sample is >2 cycles different from NCS VIC Ct, suspect PCR inhibition; retest or treat sample to remove inhibitors.

• Check expiration dates on component labels and store components under appropriate conditions (see labels).

Frequently Asked Questions (FAQ)

Q1: What is the limit of detection (LOD) of the SHENTEK MVM Detection Kit?
The reported analytical sensitivity is 50 copies per reaction.
Q2: Which MVM strains does this kit detect?
The assay is designed to detect MVM strains i, p, m and c.
Q3: What sample types are supported and what extraction is recommended?
The kit is intended for cell culture and serum samples (example: 200 µL sample input). Use the SHENTEK® Virus DNA & RNA Extraction Kit (Product No. 1506730) and rHCDpurify System for nucleic acid extraction as recommended.
Q4: How are results interpreted and what indicates inhibition?
Positive: FAM Ct < 40 (at least one well) with specific amplification and VIC Ct < 35. Negative: FAM undetected in duplicates and Ctsample - CtNCS < 2. If Ctsample - CtNCS ≥ 2 (VIC comparison), this suggests PCR inhibition; retest or remove inhibitors.
