Product Description

MicroSHENTEK® Fungi & Bacteria DNA Extraction Kit is a magnetic-particle based nucleic acid extraction kit designed to isolate trace fungal and bacterial DNA from complex matrices (≤ 10^6 cells) including master cell banks, working cell banks, vaccines and cellular therapeutic products. The kit is intended to be used together with MicroSHENTEK® Fungi DNA Detection Kit and MicroSHENTEK® Bacteria DNA Detection Kit to provide an integrated workflow from sample preparation to qPCR detection. The kit is compatible with automated extraction on the rHCDpurify system and supports inclusion of an internal control for fungal extraction. Recommended system extraction performance is 50–100 CFU per tube, and the corresponding detection kit limit of detection is ≤35 CFU/reaction.

Attention

• For research use only; read Material Safety Data Sheets (MSDS) and use appropriate PPE (eye protection, mask, gloves, lab coat).

• Separate laboratory areas for negative control/reagent prep, sample processing and positive control to avoid cross-contamination.

• Disinfect work areas with nucleic acid scavenger and 75% ethanol; perform UV irradiation of benches (≥1 hour) and work areas (≥30 minutes) before use.

• Before and after rHCDpurify runs perform UV sterilization for at least 15 minutes and wipe with 75% ethanol; minimum interval between extractions is 30 minutes.

• Maintain ambient experimental temperature ≥22 °C and humidity ≤70% during procedures.

• Change gloves and protective garments regularly and use different supplies in different areas to avoid contamination.

• Ensure magnetic particles are equilibrated and vortexed prior to use; if Lysis or Binding solutions appear cloudy, warm to 37 °C and mix until clear.

• After extraction immediately transfer purified eluate to a new tube and ensure at least 40 μL is recovered for downstream qPCR; perform qPCR the same day if possible.
