Product Description

SHENTEK® Animal-derived Biomaterial Residual DNA Sample Preparation Kit uses magnetic particle separation technology to extract residual DNA from a broad range of animal-derived biomaterials. The kit is designed for efficient and reproducible recovery of residual DNA from complex sample matrices including extracellular matrix, collagen and bio-absorbable membranes. Extracted DNA is compatible with downstream residual DNA quantitation by qPCR or PicoGreen assays. The kit supports manual sample preparation and automated extraction using the rHCDpurify system. Reagents are supplied for 50 extractions. Intended for research use only.

Attention

• Read MSDS and wear appropriate PPE (gloves, eyewear, mask, lab coat).

• Add 20 mL anhydrous ethanol to Wash buffer A before first use; prepare Wash buffer B as 70% ethanol with ddH2O.

• Prepare Proteinase K digestion solution freshly; incubate samples at 55 °C for 60 min (adjust as needed).

• Do not add Yeast tRNA when using PicoGreen DNA quantitation.

• Resuspend magnetic particles thoroughly before use; avoid over-drying pellets—dry 30 s to 3 min depending on conditions.

• Centrifuge after vortexing to collect liquid from tube cap and walls before magnetic separation.

• Transfer eluate immediately after elution and perform downstream assays the same day to ensure accuracy.

• Store magnetic particles at 2–8 °C; cold-stored reagents (e.g., glycogen) should be kept at -20 °C.

Q1: Which downstream assays are compatible with the extracted DNA?
Extracted DNA is compatible with qPCR-based residual DNA quantitation and fluorescence-based assays such as PicoGreen. Do not add Yeast tRNA if using PicoGreen.
Q2: What is the recommended digestion protocol for samples?
Prepare Proteinase K digestion solution (20 µL Proteinase K + 100 µL Proteinase K Buffer + 80 µL DEPC H2O per sample) and incubate at 55 °C for 60 minutes. Adjust enzyme amount and digestion time for difficult-to-digest or cross-linked samples.
Q3: How should Wash buffer A be prepared before first use?
Add 20 mL of anhydrous ethanol to Wash buffer A (NND015) before first use as instructed in the manual.
Q4: Can I run the kit on an automated platform?
Yes. The kit is compatible with the rHCDpurify system (program rHCD-193). Follow the plate layout and column reagent volumes specified in the manual and perform UV sterilization as recommended.
