Description

M-CSF is a disulfide-linked homodimeric glycoprotein and core member of the colony-stimulating factor family. By specifically binding the c-Fms receptor (CSF-1R/CD115), it induces receptor dimerization and internalization and activates PI3K-AKT/mTOR and Ras-MAPK signaling to drive hematopoietic stem cell differentiation toward monocyte/macrophage lineages while promoting osteoclast precursor proliferation and bone remodeling.

This protein exists as two bioactive isoforms: glycosylated and chondroitin sulfate-modified proteoglycan variants—the latter extends local bioactivity via extracellular matrix anchoring.

Immunologically, it mediates endothelial progenitor mobilization and neovascularization through macrophage-derived VEGF secretion while upregulating proinflammatory chemokines to potentiate innate immunity.

For cell therapy applications, it serves as a critical component in:

  • ex vivo macrophage differentiation systems, synergizing with GM-CSF to induce M1-polarized anti-tumor macrophages for CAR-M therapies with enhanced tumor infiltration and phagocytosis;
  • 3D bioscaffold cultures where spatiotemporal release kinetics engineer functional vascular networks to improve survival of tissue-engineered grafts.

Features

  • Native Dimeric M-CSF Activity

Disulfide-linked homodimer preserves authentic CSF-1R (c-Fms) activation and downstream signaling.

  • High Biological Potency

Verified by M-NFS-60 cell proliferation assay with specific activity ≥1×107 IU/mg (lot values up to 3.88×107 IU/mg).

  • Low Endotoxin, Stringent QC

90% purity with endotoxin <0.1 EU/µg; sterility and mycoplasma tested for sensitive cell culture use.

  • Consistent Lot Performance

Tight control of moisture and pH supports reproducible differentiation outcomes.

Applications

Macrophage Differentiation Systems

Drives monocyte/macrophage lineage commitment for in vitro immune cell studies.

CAR-M & Tumor Immunology Research

Used alone or with GM-CSF to support macrophage activation and functional polarization.

3D Tissue Engineering & Vascularization

Facilitates macrophage-mediated angiogenic support in scaffold-based cultures.

Bone & Osteoclast Research

Supports studies on osteoclast precursor proliferation and bone remodeling pathways.

Stability & Storage

  • Lyophilized powder: Stable for 12 months at −80 °C or 6 months at −20 °C when stored in the original sealed container under desiccant.
  • Reconstitution: Dissolve in sterile Water for Injection, 0.9% NaCl, or PBS (pH 7.4), maintaining a final concentration ≥100 µg/mL.
  • Handling: After reconstitution, aliquot and avoid repeated freeze–thaw cycles.

Cautions

• Research Use Only: This product is not intended for diagnostic or therapeutic use.

• Handling: Prepare solutions using sterile technique. Avoid microbial contamination to maintain activity.

• Storage: Follow recommended storage conditions. Repeated freeze–thaw cycles may reduce performance.
