Description
This product is intended for inhibiting the residual reverse transcriptase activity of DNA polymerases in PERT assays.
PERT (Polymerase Extension Reverse Transcription) is an extremely sensitive method for detecting reverse transcriptase activity. However, samples—particularly cell lysates and cell culture supernatants—often contain DNA polymerases. Notably, many prokaryotic DNA polymerases (e.g., Taq DNA polymerase) as well as eukaryotic DNA polymerases exhibit residual reverse transcriptase activity. Such activity significantly increases background signals, leading to false-positive results even in negative controls and severely interfering with the assay. It has the following features:

Ready-to-use: Add to PERT reaction system at specified ratio.

Specifically inhibits DNA polymerase's reverse transcriptase activity; no effect on most reverse transcriptases.

Compatible with all DNA polymerase types.

Inhibitory effect varies by DNA polymerase, typically ~10-fold (Ct value increases by ~3).

1 mL sufficient for 500 × 20 μL PERT reactions.

Storage and Handling
Transport at low temperature, store at -20℃, the storage period is 12 months.

Procedures
Prior to reverse transcription, add 1/10 volume of this product to the PERT reaction system (e.g., 2 μL for a 20 μL system). Follow standard protocols for the rest of the procedure. It is recommended to include a control without this product to verify efficacy. The dosage can be adjusted above or below the 1/10 volume baseline for optimization.
