Description
This kit is based on a solid-phase enzyme-linked immunosorbent assay (ELISA) using a double-antibody sandwich technique to quantify CHO PLBL-2 protein in biological samples. A CHO PLBL-2–specific capture antibody is pre-coated onto each well of the microtiter strips. Both calibration standards and test samples are simultaneously added to the wells, followed by incubation and washing. A biotinylated detection antibody is then introduced to bind an epitope of CHO PLBL-2, forming a sandwich complex that subsequently reacts with streptavidin-labeled HRP (SA-HRP). TMB (3,3′,5,5′-tetramethylbenzidine) substrate is added to the reaction and catalyzed by HRP to produce a blue-colored product (maximum absorption at 655 nm). The enzymatic reaction is terminated with the stop solution, yielding a yellow-colored product (maximum absorption at 450 nm). The absorbance at 450 nm is positively correlated with the CHO PLBL-2 concentration in both the calibration standards and the samples. The concentration of CHO PLBL-2 in the samples can be calculated using a dose–response curve.
