Preparation
Prepare a 10X solution by dissolving 6.7 g of Yeast Nitrogen Base without Amino Acids, 5 g of dextrose or an equivalent amount of another carbohydrate, and 5-10 mg of the desired amino acid in 100 mL of distilled or deionized water.
Warm slightly to dissolve and mix well. Do not bring to a boil. Do not autoclave. Filter-sterilize the solution and store the 10X stock solution at 2-8°C.
To prepare the final medium, aseptically pipette 0.5 mL of the 10X solution into 4.5 mL of sterile distilled water. Mix thoroughly by shaking before inoculation.

Storage
Store the sealed bottle containing the dehydrated medium at 2-30°C. Once opened and recapped, place the container in a low-humidity environment at the same storage temperature. Protect from moisture and light.
Discard the dehydrated medium if it is no longer free flowing or if the color has changed from the original off-white appearance.
