Description

OK432 is an immunomodulatory agent produced through the lyophilization of Group A Streptococcus pyogenes.

It activates neutrophils, enabling them to effectively kill tumor cells pretreated with IFN-γ or TNF-α. This cytotoxic effect is mediated by the interaction between CD11b/CD18 on neutrophils and ICAM-1 expressed on tumor cells.

Furthermore, OK432 stimulates monocytes to enhance their tumoricidal activity against autologous cancer cells.

In lymphocytes, OK432 induces lymphokine-activated killer (LAK) cell activity, which demonstrates potent cytotoxicity even against NK-resistant tumor cells.

Due to its immunomodulatory and antitumor properties, OK432 is suitable for the development and production of cell-based therapeutics.

OK432 is standardized in Klinische Einheit (KE), a clinical unit of biological activity. The potency is defined such that 1 KE is equivalent to 0.1 mg of the lyophilized bacterial product.

Why researchers choose OK432: 21.0% cytotoxic index (more than 2× the assay minimum) · KE-standardized potency · effective against NK-resistant tumors · sterility / mycoplasma / tumor-suppression tested · ready-to-use lyophilized format · in stock, ships in 1–2 days.

Features

  • Exceptional Potency — 21.0% Cytotoxic Index

In the standardized NK cell activity assay, OK432 drives an effector-cell cytotoxic index (T–C) of 21.0% — more than double the ≥10% assay minimum — reflecting strong, reproducible activity.

  • Standardized in KE Units for Reproducible Dosing

Activity is defined in Klinische Einheit (KE), where 1 KE = 0.1 mg lyophilized product — so you dose by biological activity, not mass, for batch-to-batch consistency.

  • Effective Against NK-Resistant Tumors

Induces lymphokine-activated killer (LAK) activity that shows cytotoxicity even against tumor lines resistant to conventional NK killing.

  • Documented COA — Three-Point Release Testing

Every lot ships with a COA covering sterility (no growth), mycoplasma (negative), and a tumor-suppression test (91% life-extension rate vs. ≥50% required), for research-grade confidence.

  • Ready-to-Use Lyophilized Powder

Supplied as a white, fluffy lyophilized powder that reconstitutes cleanly in WFI, saline, or PBS — stable for 24 months at 2–8°C.

Applications

Neutrophil Tumor Cytotoxicity

Activates neutrophils to kill tumor cells pretreated with IFN-γ or TNF-α via CD11b/CD18–ICAM-1 interaction.

Monocyte Antitumor Activation

Stimulates monocytes to enhance tumoricidal activity against autologous cancer cells.

LAK Cell Induction

Induces lymphokine-activated killer (LAK) cell activity in lymphocytes, demonstrating cytotoxicity against NK-resistant tumor cells.

NK Cell Function Studies

Used to enhance NK/LAK-mediated cytotoxicity in in vitro immune function studies

Stability & Storage

  • Lyophilized powder: Stable for 24 months at 2–8°C in the original.
  • Reconstitution: Dissolve in sterile Water for Injection, 0.9% NaCl, or PBS (pH 7.4) maintaining final concentration ≥100 µg/mL to prevent adsorption.
  • Handling: Aliquot to avoid repeated freeze-thaw cycles.

Cautions

• Research Use Only: This product is not intended for diagnostic or therapeutic use.

• Handling: Prepare solutions using sterile technique. Avoid microbial contamination to maintain activity.

• Storage: Follow recommended storage conditions. Repeated freeze–thaw cycles may reduce performance.

FAQ

What is OK432 (Picibanil) used for in immunology research?

OK432 is used as an innate immune activation reagent for NK/LAK activation, macrophage stimulation, and dendritic cell maturation in tumor immunology and immunotherapy research models.

What grade is this, and can I use it clinically?

This product is supplied for Research Use Only (RUO) and process-development use. It is not sold for diagnostic or therapeutic use, and nothing here is a therapeutic claim. For specification, grade, or documentation questions, contact us through biofargo.com.

Is OK432 a TLR agonist?

OK432 is a whole-bacterial immunomodulator reported to engage TLR-associated innate pathways (notably TLR4), contributing to cytokine release and downstream immune cell activation.

How does OK432 activate NK cells?

OK432 does not bind NK cells directly. It stimulates accessory cells to release NK-activating factor (NKAF) and other cytokines, which in turn enhance NK cytotoxicity, and it induces lymphokine-activated killer (LAK) activity effective even against NK-resistant targets.

Which immune cells respond to OK432 stimulation?

OK432 activates NK cells, monocytes, macrophages, neutrophils, and dendritic cells, making it widely used in innate immunity and tumor microenvironment studies.

Can OK432 be used in dendritic cell maturation studies?

Yes. OK432 is frequently used to promote dendritic cell maturation and antigen presentation in immunotherapy and adaptive immunity research workflows.

What are the storage conditions for OK432?

Store at 2–8°C in a light-protected environment. After reconstitution, aliquot and avoid repeated freeze–thaw cycles to maintain biological activity.

Is OK432 RUO or clinically approved?

OK432 is strictly labeled as Research Use Only (RUO) and is not intended for human or animal therapeutic use.

When can I expect my order to ship?

Our standard shipping usually take 2-5 days.

We also provide express shippping for time-sensitive deliveries.
