Description

Tryptic Soy Agar (TSA) is a general-purpose culture medium used for the cultivation and isolation of both fastidious and non-fastidious microorganisms, as well as for maintaining stock cultures. It is commonly utilized in the pre-cultivation and enumeration of E. coli according to the membrane filtration technique. TSA supports the growth of both aerobes and anaerobes.

To prepare blood agar, 5–10% sterile defibrinated blood can be added. TSA supplemented with 5% sheep blood is particularly effective for isolating and identifying hemolytic reactions in Staphylococcus spp., Streptococcus spp., and other pathogenic microorganisms found in clinical specimens such as respiratory tract secretions, blood, cerebrospinal fluid, and minor lesions.

Formula per Liter:

• Tryptone 15.0g

• Soy Peptone 5.0g

• Sodium chloride 5.0g

• Agar 15.0g

• Final pH: 7.3 ± 0.2 at 25.0°C
