Description

FREY agar is a nutritionally enriched, semi-selective solid medium designed for the cultivation and detection of avian mycoplasmas. The formulation is supplemented with nicotinamide adenine dinucleotide (NAD⁺), a critical coenzyme required for the growth of NAD-dependent species such as Mycoplasma synoviae.

The presence of phenol red as a pH indicator enables visual monitoring of metabolic activity, with a color shift from red to yellow (acidic) or purple (alkaline) indicating the proliferation of mycoplasmal organisms. To selectively inhibit contaminating bacterial flora while preserving mycoplasma viability, penicillin G is incorporated into the medium.

Furthermore, the addition of exogenous lipid supplements from yolk, excluding antibiotics—including cholesterol and long-chain fatty acids—enhances the growth performance of lipid-requiring strains, particularly Mycoplasma pneumoniae ATCC 23714, by supporting membrane biosynthesis and promoting optimal colony formation.

Application

FREY agar is recommended for the detection of avian mycoplasmas in pharmaceutical products, particularly in bulk vaccines, cell banks, and virus cultures.

Features

  • Ready-to-use convenience — no additional preparation steps are required.

  • Broad pharmaceutical applications — suitable for biopharmaceuticals, vaccines, cell cultures, and virus cultures.

  • Storage stability — can be stored at 2–8 °C for up to 3 months.

  • Rapid detection — a colorimetric chart provides a quick and convenient visual indication of early-stage mycoplasma proliferation.

  • Optimized growth performance — the buffered system supports higher colony counts while reducing the risk of mycoplasma cell death.

  • Enhanced visualization — a chromogenic reagent selectively stains mycoplasma, improving clarity and specificity of detection under microscopy.

  • Improved NAD⁺ stability — to prevent growth inhibition caused by NAD⁺ degradation at later stages, alternative NAD⁺ analogs are co-supplemented in the culture system.
