Product Description
SHENTEK® Residual Porcine DNA Quantitation Kit is designed to quantify residual porcine DNA in biomaterials using fluorescent quantitative PCR (qPCR). The kit provides a rapid, specific and reliable absolute quantitation assay with sensitivity down to the femtogram (fg) level. The product is supplied for 100 reactions (Product No. 1101113) and is intended for research use only. For sample extraction instructions, refer to the SHENTEK® Animal-derived Biomaterial Residual DNA Sample Preparation Kit (Product No. 1104193). The kit includes internal positive control (IPC) reagents and a porcine DNA standard control to build a standard curve for absolute quantitation.
Technical Specifications
| Parameter | Details |
|---|---|
| Intended use | Quantitation of residual porcine DNA in biomaterials (research use only) |
| Sensitivity | Detectable at femtogram (fg) level (kit optimized for absolute quantitation down to 0.03 pg/µL standard point) |
| Standard curve range (prepared) | Serial standards prepared: ST0 30000 pg/µL (stock), ST1 3000 pg/µL, ST2 300 pg/µL, ST3 30 pg/µL, ST4 3 pg/µL, ST5 0.3 pg/µL, ST6 0.03 pg/µL |
| Reaction volume | Total reaction volume 30 µL (20 µL qPCR MIX + 10 µL sample/standard) |
| qPCR MIX composition (per reaction) | AT qPCR Reaction Buffer 15.9 µL; Porcine Primer & Probe MIX 2.8 µL; IPC MIX 1.3 µL; (optional UNG enzyme 0.1 U) |
| Thermal cycling conditions | Optional UNG 25 °C 10:00; Activation 95 °C 10:00; 40 cycles of 95 °C 00:15 (denaturation) and 60 °C 00:40 (annealing/extension, fluorescence read) |

Features
- High sensitivity: Optimized qPCR assay capable of femtogram-level detection for low-abundance residual porcine DNA.
- Absolute quantitation: Includes porcine DNA control and recommended serial dilutions to generate an absolute-quantitation standard curve.
- Internal control (IPC): IPC MIX included to monitor extraction and PCR inhibition for each sample.
- Ready-to-use mixes: Pre-formulated AT qPCR Reaction Buffer and Porcine Primer & Probe MIX reduce setup time and variability.
- Carryover prevention option: Supports optional UNG treatment step when UNG enzyme is included to prevent carryover contamination.
- Validated workflow: Detailed guidance for serial dilution, plate layout, thermal cycling and result analysis for reproducible assays.
Applications
- Quantitation of residual porcine DNA in biomaterials (e.g., tissue-derived products, implants, scaffolds)
- Quality control and release testing for animal-derived material processing
- Method validation and extraction recovery assessment using ERC samples
- Research studies requiring sensitive detection of porcine DNA contamination
Kit Contents
Attention
• For Research Use Only (RUO); not for diagnostic use.
• Read Material Safety Data Sheets (MSDS) and follow handling instructions. Wear appropriate PPE (eyewear, mask, clothing, gloves).
• Decontaminate work surfaces, pipettes and tubes by UV irradiation (30 minutes) and 75% ethanol prior to setup to minimize contamination.
• Thaw kit reagents at 2–8 °C or on ice, vortex and briefly centrifuge before use.
• Store reagents at –20 °C; check expiration dates on labels. Protect light-sensitive reagents.
• Include at least five standard concentrations when preparing the standard curve; recommended serial dilution series provided (ST1–ST6).
• Prepare NTC, NCS and ERC controls as instructed. Test samples and controls in triplicate for reliable results.
• If UNG is used, include 25 °C 10:00 UNG step in the thermal program; UNG is optional but recommended to prevent carryover.
• Monitor IPC Ct values to assess inhibition; acceptable IPC Ct shift is ±1.0 relative to NCS.
• Acceptable ERC recovery rate is 50%–150%; if outside this range investigate extraction and inhibition.
