Product Description

This is a sandwich ELISA kit for quantitative detection of Human Interferon Gamma (IFN-γ). Microtiter strips are pre-coated with an affinity-purified anti-Human IFN-γ capture antibody. Samples and calibration standards are incubated in the wells, followed by binding of a biotinylated detection antibody and detection with streptavidin–HRP. TMB substrate is oxidized by HRP to produce a blue product (max absorbance ~655 nm) that is converted to yellow upon addition of stop solution (read at 450 nm). The assay is calibrated against recombinant human IFN-γ (NIBSC/WHO reference evaluated) and is intended for research use (e.g., potency assays for CAR-T cells and measurement in cell culture supernatants).

• Avoid repeated freeze–thaw cycles of standards and samples; long‑term storage at -65 °C or below recommended for samples.

• Dissolved Calibration Standards, 1× Biotinylated Conjugate and 1× Streptavidin-HRP are recommended for single use (prepare freshly).

• Do not mix reagents from different lot numbers.

• Use fresh sterile or ultra‑pure water; avoid water >37 °C.

• Avoid samples containing sodium azide (NaN3) which will inhibit HRP and cause underestimation.

• Stop Solution is 1 M HCl — avoid direct contact with eyes, skin and clothing; wear goggles and protective equipment.

• TMB Substrate should be colorless before use; discard if colored.

• Seal microplate during incubations to prevent evaporation; avoid drying wells before substrate addition.

• Centrifuge conjugate reagents (100×) before dilution to avoid loss of reagent.

• Minimum recommended pipetting volume for dilution accuracy: 5 μL.

• Complete plate reading within 30 minutes after termination.

Q4: What should I do if my substrate is colored or background is high?
If TMB substrate is colored, discard and contact technical support. For high background, review washing steps, avoid cross-contamination, clean equipment and prepare fresh reagents.
