Product Description

The SHENTEK® BPV-1 Analysis Kit is a real-time PCR based qualitative assay serving as an analytical quality control reagent engineered exclusively for the molecular evaluation of bioprocessing inputs, such as in vitro cell culture supplements and cell banks. Optimized for compatibility with the SHENTEK® Nucleic Acid Extraction Kit (Product No. 1506730), this assay ensures the efficient recovery and analysis of target nucleic acids from complex industrial matrices.

Reaction setup 20 µL qPCR MIX + 10 µL template (extracted sample / control / DDB); final reaction volume 30 µL
qPCR mix composition (per reaction) BPV-1 Primer&Probe MIX 2.8 µL; qPCR Reaction Buffer 15.9 µL; IPC MIX 1.3 µL; Total 20 µL
Thermal cycling conditions Activation: 95 °C 10:00 (1 cycle); Denaturation: 95 °C 00:15 (45 cycles); Annealing/Extension: 60 °C 01:00 (instrument reads fluorescence during this step)
Controls and interpretation thresholds (FAM / VIC) NTC: Ct ≥ 40.00 or no amplification (FAM) / Ct < 35.00 for VIC indicates valid no-template control; NCS: Ct ≥ 40.00 or no amplification (FAM) / Ct < 35.00 (VIC); PCS: Ct < 35.00 (FAM and VIC); Test sample interpretation: Positive if FAM Ct < 40.00 (at least one well) with specific amplification and VIC Ct < 40.00 for duplicate runs; Presumptive positive if FAM Ct ≥ 40.00 or absent and VIC Ct < 40.00 indicating possible inhibitors, etc. See user guide tables for complete criteria.
Compatible instruments SHENTEK-96S, LightCycler 480 II, ABI 7500, CFX96 and other standard real-time PCR systems supporting probe assays
Storage stability / Shelf life Kit components can be stored under appropriate conditions up to 24 months (see labels)
Extraction compatibility Validated for use with SHENTEK® Virus DNA & RNA Extraction Kit (Product No. 1506730)

Features

  • High sensitivity: Limit of detection down to 50 copies per reaction enables detection of low-level BPV-1 contamination.
  • High specificity: No cross-amplification with bovine/porcine genomes, common mammalian cell lines (CHO, Vero, 293T, MDCK, NS0, Sf9) or common expression hosts (E. coli, Pichia pastoris).
  • Complete qPCR solution: Includes primer/probe mix, qPCR reaction buffer and an internal positive control (IPC) to monitor inhibition.
  • Designed for complex sample matrices: Works with SHENTEK® Virus DNA & RNA Extraction Kit to recover and detect viral DNA from cells and serum.
  • Broad instrument compatibility: Compatible with major real-time PCR platforms (SHENTEK-96S, LightCycler 480 II, ABI 7500, CFX96).

Applications

  • Qualitative molecular analysis of BPV-1 target sequences in industrial cell banks
  • Analytical quality control of biological seed lots
  • Molecular evaluation of in vitro cell culture supplements for BPV-1 nucleic acids
  • In-process monitoring and lot-release analysis for biological manufacturing
  • In vitro research projects assessing BPV-1 target sequences in complex culture matrices

Attention

• For Research Use Only (RUO). Not for diagnostic or clinical use.

• Read Material Safety Data Sheets (MSDS) and follow handling instructions; wear appropriate PPE (protective eyewear, mask, clothing and gloves).

• Handle positive control and extraction procedures in a biosafety cabinet and follow BSL-2 laboratory safety guidelines.

• Use nuclease-free consumables and reagents certified as RNase/DNase-free.

• Thaw kit components at 2–8 °C or on ice; vortex and centrifuge briefly before use.

• Protect primer/probe mix from light.

• If VIC (internal control) signal indicates inhibition, repeat sample preparation or remove inhibitors and repeat the assay.

Frequently Asked Questions (FAQ)

Q1: What is the limit of detection (LOD) for this kit?
The assay sensitivity reaches 50 copies per reaction (LOD = 50 copies/reaction).
Q2: Which sample types are supported?
The kit is validated for cell culture samples (≤10^7 cells) and bovine serum. Other sample types may be processed after appropriate extraction; consult technical support for special matrices.
Q3: What controls should be included in each run?
Include at minimum: No Template Control (NTC), Negative Control Sample (NCS), and Positive Control Sample (PCS). The guide recommends running samples in duplicate.
Q4: What should I do if the internal control (VIC) is inhibited?
If VIC signal is inhibited, repeat sample preparation or perform additional inhibitor removal steps, then re-run the assay. Persistent inhibition may require re-extraction or dilution of the nucleic acid.
