Product Description

The SHENTEK® Bovine Virus Analysis Multiplex Kit (BVDV/REO-3/PI-3/BPV-3/BAV-3) is a multiplex real-time PCR based qualitative assay serving as an analytical quality control reagent engineered exclusively for the simultaneous molecular evaluation of bioprocessing inputs, such as in vitro cell culture supplements and cell banks. Optimized for compatibility with the SHENTEK® Nucleic Acid Extraction Kit (Product No. 1506730), this assay ensures the efficient recovery and multiplexed analysis of target nucleic acids from complex industrial matrices.

Technical Specifications

Parameter Details
Limit of Detection (LOD) 50 copies per reaction

Thermal cycling (qPCR for BPV-3/BAV-3) Activation 95 °C 10:00 (1 cycle); 40 cycles of Denaturation 95 °C 00:15 and Annealing/Extension 60 °C 01:00 (fluorescence read during annealing/extension).
Reporter dyes / Targets REO-3: FAM; PI-3: HEX; BVDV: CY5; BPV-3: FAM; BAV-3: CY5; Internal Control (IC): ROX.
Acceptance criteria — controls NTC: Undetected (no specific amplification); NCS: Undetected; PCS: Ct < 35.00 with specific amplification; ROX (IC) for controls: Ct < 35.00.
Acceptance criteria — test samples Positive call: specific amplification with Ct < 40.00 in the relevant fluorescence channel. Sample inhibition check: compare sample ROX Ct to NCS ROX Ct; difference > 2 indicates possible inhibition. If Ctsample - CtNCS < 2 then negative; ≥ 2 not conclusive (PCR inhibitors possible).
Instrument compatibility SHENTEK-96S Real-time PCR System, LightCycler 480Ⅱ, ABI 7500 (other real-time PCR instruments may be used with appropriate setup).
Controls included / recommended Internal Control (VIR IC) provided in kit; Positive Controls for individual targets sold separately (Product Nos. 1506736, 1506737, 1506738, 1506746, 1506739).

Features

  • Multiplex detection: Simultaneous detection of three RNA viruses (BVDV/REO-3/PI-3) or two DNA viruses (BPV-3/BAV-3) in a single well using multiplex real-time PCR.
  • High sensitivity: Limit of detection down to 50 copies per reaction.
  • Internal control provided: VIR Internal Control (IC) included to monitor extraction and amplification performance and identify inhibition.
  • Compatible with standard instruments: Validated on SHENTEK-96S, LightCycler 480Ⅱ and ABI 7500; adaptable to other real-time PCR platforms.
  • Optimized workflow for complex matrices: Designed to work with SHENTEK® Virus DNA & RNA Extraction Kit for efficient recovery from serum and cell matrices.

Attention

• For Research Use Only (RUO). Not for diagnostic or clinical use.

• Read Material Safety Data Sheets (MSDS) and follow handling instructions.

• Wear appropriate PPE: protective eyewear, mask, lab coat and gloves.

• Handle positive controls and extraction procedures in a biological safety cabinet following BSL-2 guidelines.

• Use nuclease-free consumables and reagents certified RNase/DNase-free.

• Thaw kit components completely at 2–8 °C or on ice, vortex thoroughly and centrifuge briefly before use.

• Protect probe-containing reagents from light.

• Check control acceptance criteria (NTC/NCS/PCS/IC Ct thresholds) for run validity.

• If ROX (IC) Ct(sample) - ROX Ct(NCS) > 2, investigate potential PCR inhibition and repeat extraction/dilution as needed.

Frequently Asked Questions (FAQ)

Q1: What sample types are compatible with this kit?
The kit is validated for bovine serum and cell culture matrices (cell banks). Use SHENTEK® Virus DNA & RNA Extraction Kit for nucleic acid extraction prior to detection.
Q2: What is the assay sensitivity (LOD)?
The assay has a reported limit of detection of 50 copies per reaction.
Q3: What controls should be included in each run?
Include No Template Control (NTC), Negative Control Sample (NCS), and Positive Control Sample (PCS). An internal control (VIR IC) is provided to monitor extraction and amplification; add IC during extraction or to the reaction mix if not already added.
Q4: How are positive/negative results determined?
A positive result is a well showing specific amplification with Ct < 40. For controls, PCS and ROX (IC) should have Ct < 35. Compare ROX Ct of sample to NCS to assess inhibition (difference > 2 suggests inhibition).
