Description

iPSC-Derived Vascularized Lung Organoid Differentiation Kit is a chemically defined, directed-differentiation system that efficiently converts human pluripotent stem cells (hPSCs), including iPSCs, into multi-lineage vascularized lung organoids through three induction stages: mesendoderm, vascularized primitive gut, and vascularized lung organoid.

The resulting organoids show multi-level branching and early alveolus-like structures. They contain several physiologically relevant alveolar and airway cell lineages together with vascular endothelial cells, giving a lung model with an integrated vascular component. One kit supports the differentiation of 48 organoids.

Features

  • Vascularized lung organoids containing both lung epithelial and vascular endothelial lineages.

  • Multi-level branching and early alveolus-like structures.

  • Alveolar and airway lineages (e.g., SPC-positive alveolar type II and KRT5-positive basal cells) with NKX2.1 lung identity.

  • Chemically defined, stage-specific media — each step has its own ready-to-use medium.

  • Sufficient for 48 organoids per kit.

Applications

  • Lung Development: Modeling branching morphogenesis and early alveolar development.

  • Lung Disease Modeling: Fibrosis, infection and injury models that involve the epithelial–vascular interface.

  • Respiratory Virus Research: Human lung tissue models for infection studies.

  • Drug and Toxicity Testing: 3D human lung models with a vascular compartment.

Protocol Overview

  1. Begin when iPSCs reach about 80% confluence. Dissociate to single cells and culture for 24 hours in iPSC medium with 10 µM Y-27632 in a 96-well ultra-low-attachment U-bottom plate to form embryoid bodies.

  2. Day 0–3 (mesendoderm): change the medium daily — Medium A on Day 0–1, Medium B on Day 1–2, Medium C on Day 2–3.

  3. Day 3–7 (vascularized primitive gut): switch to Medium D and change daily.

  4. Day 7: transfer organoids onto a basement membrane matrix-coated Transwell insert and add Medium E to the lower chamber; change every other day until Day 10.

  5. Day 10–26: switch to Medium F and change every other day until the organoids mature.

Frequently Asked Questions (FAQs)

Common questions about iPSC-Derived Vascularized Lung Organoid Differentiation Kit.

What cells are used as the starting material?

Human pluripotent stem cells, including iPSCs, at about 80% confluence.

How long does differentiation take?

About 26 days to maturation, with culture continuing beyond Day 30 if needed.

How many organoids does one kit make?

One kit supports the differentiation of 48 vascularized lung organoids.

Which markers can be used to characterize the organoids?

Lung identity and lineage markers such as NKX2.1, SPC and KRT5, and VEGF for the vascular component.

Can it be used for clinical or diagnostic purposes?

No. This product is for research use only and is not intended for human, diagnostic or therapeutic use.
