Introduction
Reliable separation and transfer of high-molecular-weight (HMW) proteins is often a major challenge for researchers. While 4–20% Tris-Glycine gels are commonly used for a broad range of proteins, large proteins tend to remain compressed near the top of the gel. For superior performance, Tris-Acetate gels provide a better solution. Operating at a near-neutral pH (~8.1), Tris-Acetate gels deliver enhanced resolution and improved migration for HMW proteins compared to Bis-Tris or Tris-Glycine systems. The lower acrylamide concentration facilitates smoother protein movement, resulting in higher transfer efficiency and greater detection sensitivity. Moreover, the neutral buffer environment minimizes protein modification, producing cleaner, sharper bands ideal for SDS-PAGE and Western blot applications.

It enables effective separation of large protein molecules (35–3 kDa) with a short electrophoresis time. At 150 V, electrophoresis can be completed in approximately 6 minutes. The gel does not contain SDS, allowing it to be used for both denaturing and native electrophoresis, and it is compatible with most mini electrophoresis tanks on the market. |
