Description

Traditional SDS-PAGE gel preparation requires multiple manual procedures, including precise acrylamide concentration adjustment, separate preparation of resolving and stacking gels, polymerization reagent handling, and repeated optimization for different protein sample types. These steps can increase preparation time and introduce variation, especially when high-resolution separation of small proteins or peptides is required.

The 15% One-Step SDS-PAGE Color Gel Fast Preparation Kit (RP0261) is a complete SDS-PAGE gel preparation kit designed for high-resolution separation of low molecular weight proteins and peptide samples using polyacrylamide gel electrophoresis. The kit provides pre-optimized separation gel solution, concentrating gel solution, corresponding buffers, and a novel coagulant system, allowing researchers to prepare PAGE gels without additional TEMED addition or complex reagent calculations.

Designed for the Tris-glycine SDS-PAGE system, the RP0261 One-Step PAGE Gel Kit contains a 15% polyacrylamide gel formulation with smaller pore sizes for improved resolution of small proteins compared with lower percentage SDS-PAGE gels. It is particularly suitable for applications requiring enhanced separation of proteins below approximately 50 kDa, including detailed protein characterization and high-resolution Western blot analysis.

The colored concentrating gel included in this protein gel casting kit improves visualization of sample loading wells and supports accurate sample application during electrophoresis. Compared with conventional hand-cast SDS-PAGE methods, the TEMED-free SDS-PAGE gel preparation workflow reduces preparation complexity and improves consistency for laboratories performing routine and advanced protein analysis.

The RP0261 kit is intended for research use only and is suitable for applications including small protein separation, peptide electrophoresis, Western blot gel preparation, protein expression analysis, and polyacrylamide gel electrophoresis.

Applications

The 15% One-Step SDS-PAGE Color Gel Fast Preparation Kit is designed for laboratories requiring high-resolution separation of small proteins and peptide samples using polyacrylamide gel electrophoresis.

  • Small protein separation: Suitable for resolving low molecular weight proteins that require higher percentage SDS-PAGE gels for improved separation.

  • Peptide electrophoresis: Provides enhanced resolution for peptide samples and very small protein fragments.

  • Western blot analysis: Preparation of SDS-PAGE gels for detection of low molecular weight proteins before membrane transfer.

  • Protein characterization: Analysis of purified proteins, protein fragments, and molecular weight differences.

  • Protein expression analysis: Evaluation of recombinant proteins and engineered protein products with lower molecular weights.

  • Biotechnology and academic research: Convenient high-resolution SDS-PAGE gel preparation for molecular biology laboratories.

Choosing the Right SDS-PAGE Gel Percentage

The optimal SDS-PAGE gel concentration depends on protein molecular weight and the required separation resolution. Lower percentage polyacrylamide gels provide larger pores for high molecular weight proteins, while higher percentage gels provide improved resolution for smaller proteins. The 15% SDS-PAGE gel contains a higher acrylamide concentration and smaller pore structure, making it suitable for high-resolution separation of small proteins, protein fragments, and peptide samples.

Gel Concentration Recommended Application Typical Protein Range
6% SDS-PAGE Gel High molecular weight proteins and large protein complexes >100 kDa proteins
8% SDS-PAGE Gel Medium to high molecular weight protein separation Approximately 50–200 kDa proteins
10% SDS-PAGE Gel General protein electrophoresis and routine Western blot workflows Approximately 20–150 kDa proteins
12% SDS-PAGE Gel Routine protein separation with improved resolution for smaller proteins Approximately 10–100 kDa proteins
15% SDS-PAGE Gel Small protein, peptide, and low molecular weight protein analysis <50 kDa proteins

Preparation Workflow

The RP0261 One-Step SDS-PAGE Gel Preparation Kit provides a simplified workflow for preparing high-resolution protein electrophoresis gels. The pre-optimized formulation reduces manual preparation steps while maintaining compatibility with standard Tris-glycine SDS-PAGE systems.

  1. Mix equal volumes of Separation Gel Solution and Separation Gel Buffer to prepare the resolving gel mixture.

  2. Add Novel Coagulant to the separation gel solution and gently mix to initiate polymerization while avoiding bubble formation.

  3. Transfer the prepared resolving gel solution into the gel cassette.

  4. Mix Concentrating Gel Solution and Concentrating Gel Buffer to prepare the stacking gel solution.

  5. Add Novel Coagulant and directly cast the concentrating gel above the separation gel layer without waiting for complete resolving gel polymerization.

  6. Insert the comb and allow approximately 15 minutes for gel polymerization before performing SDS-PAGE electrophoresis.

The simplified casting procedure reduces preparation complexity and improves reproducibility for laboratories performing high-resolution separation of small proteins and peptide samples.

Technical Notes

  • The RP0261 kit is designed for denaturing SDS-PAGE electrophoresis. Separation and concentrating gel buffers contain SDS and are not intended for native PAGE applications.

  • The 15% polyacrylamide formulation provides smaller pore sizes than lower percentage SDS-PAGE gels, making it suitable for enhanced resolution of low molecular weight proteins.

  • The colored concentrating gel improves visualization of sample loading wells and supports accurate sample application during electrophoresis.

  • High-percentage SDS-PAGE gels may provide improved separation of small proteins but may require optimization of electrophoresis conditions depending on sample properties.

  • Gel polymerization speed is temperature dependent. Lower temperatures may extend polymerization time, while suitable room temperature conditions support efficient gel formation.

  • For optimal electrophoresis performance, freshly prepared gels are recommended for use.

FAQ

What is a 15% SDS-PAGE Gel Preparation Kit used for?

A 15% SDS-PAGE Gel Preparation Kit is designed for high-resolution separation of small proteins, protein fragments, and peptide samples. The RP0261 kit provides a high-percentage polyacrylamide gel formulation suitable for applications requiring improved resolution of low molecular weight proteins.

Why choose a 15% SDS-PAGE gel instead of a 12% gel?

A 15% SDS-PAGE gel contains a higher acrylamide concentration and smaller pores compared with a 12% gel. This provides improved separation capability for smaller proteins and peptide samples that may not be sufficiently resolved using lower percentage gels.

Can RP0261 be used for Western blot analysis?

Yes. RP0261 can be used for Western blot workflows requiring separation of low molecular weight proteins before membrane transfer and antibody-based detection.

What proteins are suitable for a 15% SDS-PAGE gel?

The RP0261 15% SDS-PAGE gel is recommended for small proteins, protein fragments, and peptide samples generally below approximately 50 kDa. Actual separation performance depends on electrophoresis conditions and sample characteristics.

Does RP0261 require additional TEMED during gel preparation?

No. RP0261 uses an optimized coagulant system that simplifies SDS-PAGE gel preparation without requiring additional TEMED addition during the casting process.

What are the advantages of a high-percentage SDS-PAGE gel?

High-percentage SDS-PAGE gels provide smaller pore sizes, allowing improved separation resolution for lower molecular weight proteins and peptides compared with lower percentage polyacrylamide gels.

Is RP0261 compatible with standard SDS-PAGE electrophoresis equipment?

Yes. RP0261 is designed for standard Tris-glycine SDS-PAGE systems and can be used with conventional PAGE gel casting and electrophoresis equipment.

What is the difference between RP0261 15% gel and RP0251 12% gel?

RP0251 12% SDS-PAGE gel provides routine protein separation with improved resolution for many commonly analyzed proteins, while RP0261 15% SDS-PAGE gel is optimized for smaller proteins and peptide samples requiring higher resolution.
