The World Health Organization (WHO), the U.S. Food and Drug Administration (FDA), the European Pharmacopoeia, and the Chinese Pharmacopoeia have all established regulations requiring identification tests for production cell lines to confirm their origin and to detect any contamination from other cell lines. The SHENTEK® 10-Species Cell Line Authentification kit utilizes mitochondrial genes cytochrome oxidase I (COI), cytochrome B (Cyt B), and cytochrome oxidase II (COII) as molecular identification targets for the 10 commonly used cell lines in biological product manufacturing. It employs multiplex PCR technology to identify species and detect cross-contamination based on the presence or absence of PCR amplification and the size of PCR amplicons for the given 10 sets of primers.
The 10 species of cell lines identified by this kit are as follows: Bos taurus (Cattle) / Mus musculus (Mouse) / Canis familiaris (Dog) / Rattus norvegicus (Rat) / Cercopithecus aethiops (African green monkey) / Macaca mulatta (Rhesus monkey) / Cricetulus griseus (Chinese hamster) / Felis catus (Cat) / Homo sapiens (Human) / Sus scrofa (Pig).
