Description

Feeder-free culture of human pluripotent stem cells depends on a consistent attachment surface. Variation in the coating matrix can affect colony attachment, morphology and expansion from one passage to the next.

Biofargo Basement Membrane Matrix, iPSC-Qualified is a basement membrane extract prepared from mouse sarcoma tissue and supplied at a protein concentration of 8–12 mg/mL. It is intended for thin-layer coating of culture vessels for feeder-free culture and expansion of human embryonic stem cells (hESCs) and induced pluripotent stem cells (iPSCs).

The matrix is liquid at 2–8°C and gels rapidly above 10°C. Its temperature-dependent gelation is reversible, so it can be thawed, aliquoted and diluted on ice before coating.

Features

iPSC-Qualified — Supplied for feeder-free culture and expansion of human iPSCs and ESCs.
8–12 mg/mL Protein Concentration — Basement membrane extract suitable for dilution to a thin-layer coating.
Reversible Temperature-Dependent Gelation — Liquid at 2–8°C and gels above 10°C; prematurely gelled matrix can be returned to a liquid state at 4°C.
Long Shelf Life — 24 months when stored at −80°C.
Aliquot-Friendly 5 mL Format — Thaw once, aliquot by intended use, and avoid repeated freeze–thaw cycles.

Instructions for Use

Thaw the matrix overnight at 2–8°C. For best temperature control, place the vial on wet ice inside a 4°C refrigerator during thawing.
Keep the thawed matrix on ice throughout handling. The matrix gels rapidly above 10°C.
Choose the coating thickness and matrix concentration for your application. For feeder-free culture and expansion of hESCs and iPSCs, use a thin-layer coating, not a thick gel.

Handling Notes

Store at −80°C for long-term storage. Do not use matrix that has been kept at 4°C for more than 24 hours.
After thawing, aliquot into sterile tubes according to intended use. Avoid repeated freeze–thaw cycles.
If the matrix gels prematurely, incubate at 4°C for 12–24 hours to restore the liquid state.
If the matrix looks uneven after thawing, pipette gently up and down until homogeneous.
To remove air bubbles, centrifuge at 5,000 × g for 1 minute at 0–4°C.
Follow appropriate laboratory biosafety procedures and wear suitable personal protective equipment.

Applications

Feeder-free hESC and iPSC culture — Thin-layer coating of plates and flasks for routine pluripotent stem cell maintenance.
iPSC expansion — Attachment surface for scaling up human iPSC cultures.
iPSC differentiation workflows — Coated surface for pluripotent stem cells before directed differentiation.

FAQ

What is this matrix used for?

It is used as a thin-layer coating for feeder-free culture and expansion of human ESCs and iPSCs.

What is the protein concentration?

8–12 mg/mL.

How should it be stored?

Store at −80°C. Shelf life is 24 months. Do not use matrix kept at 4°C for more than 24 hours.

What if the matrix gels before I use it?

Incubate at 4°C for 12–24 hours to return it to a liquid state.

Should I use a thin layer or a thick gel?

For feeder-free hESC/iPSC culture and expansion, use a thin-layer coating rather than a thick gel.

Is it for clinical use?

No. For Research Use Only. Not intended for clinical use.
