Description

Human NK cells can be particularly sensitive to freezing and thawing. Loss of membrane integrity, reduced post-thaw viability and incomplete cell recovery may disrupt downstream expansion, phenotyping and functional assays. These challenges become more significant when laboratories need to freeze highly concentrated NK cell preparations, transport frozen samples on dry ice or establish reproducible long-term cell banks.

Biofargo NKCryo is a serum-free NK cell cryopreservation medium developed specifically for human natural killer cell preservation. Its formulation provides cryoprotective components and nutritional support intended to reduce cellular stress during freezing, dry ice transportation and thawing while helping preserve cell viability and recovery after storage.

This human NK cell freezing medium contains no human- or animal-derived ingredients and no protein components, helping reduce the introduction of heterogeneous biological materials into frozen cell preparations. The formulation contains 10% DMSO and uses cell culture-grade, low-endotoxin raw materials.

Biofargo NKCryo supports a broad freezing-density range of 1 × 10⁶ to 5 × 10⁸ cells/mL. It is suitable for human NK cells, including NK cells derived from peripheral blood mononuclear cells and cord blood mononuclear cells, and can be integrated into NK cell expansion, cryopreservation and post-thaw recovery workflows.

Research use only: Biofargo NKCryo is intended for laboratory research applications and is not intended for direct therapeutic or diagnostic use.

Performance Data

Post-Thaw NK Cell Viability After Cryopreservation

Cryopreservation performance was evaluated using human NK cells with a CD3^-CD56⁺ proportion greater than 90%. Cells were generated from cord blood-derived mononuclear cells and expanded using a Biofargo NK activation workflow before freezing with Biofargo NKCryo.

The evaluation demonstrated that Biofargo NKCryo supports high NK cell survival after cryopreservation. Under high-density freezing conditions, cells maintained post-thaw viability above 90% after 3 months of storage and maintained viability above 80% during a 6-month storage period.

Recommended Cryopreservation Workflow

Biofargo NKCryo provides a standardized workflow for laboratories performing human NK cell expansion, freezing and recovery experiments. The recommended process includes cell harvesting, cryoprotectant addition, controlled-rate freezing and transfer to long-term storage.

Step 1: NK Cell Collection

After completion of NK cell culture and expansion, collect NK cells by centrifugation and carefully remove the culture supernatant.

Step 2: Add Biofargo NKCryo Freezing Medium

Slowly add Biofargo NKCryo cryopreservation medium to the cell pellet. Gently resuspend the cells to achieve a uniform suspension.

Recommended final freezing density:

1 × 10⁶ to 5 × 10⁸ cells/mL

Step 3: Controlled Cooling and Storage

Transfer prepared cryovials into a controlled-rate freezing container and place them at -80°C. For long-term storage, transfer cells to liquid nitrogen storage after approximately 12 hours.

Step 4: NK Cell Thawing and Recovery

Thaw frozen NK cells using a 37°C water bath or cell recovery instrument. Transfer cells into a centrifuge tube and add at least three volumes of serum-free NK cell culture medium or physiological saline to dilute the cryopreservation solution. Centrifuge to remove the freezing medium before downstream experiments.
