Description

Maintaining reliable human mesenchymal stem cell expansion can be difficult when serum- or platelet lysate-based culture systems introduce lot-to-lot variability, inconsistent proliferation, or changes in cell characteristics during serial passage. Laboratories working with umbilical cord, bone marrow, or adipose-derived MSCs need a more standardized culture environment that supports cell yield without compromising multipotency.

Biofargo MSC Max Kit is a serum-free human mesenchymal stem cell culture kit designed for the isolation and continuous expansion of human MSCs. The kit combines a dedicated basal medium with an optimized supplement to provide a complete human MSC serum-free medium for routine propagation and long-term culture.

The animal component-free formulation provides an alternative to conventional αMEM plus human platelet lysate culture workflows. Under the evaluated conditions, the medium supported repeated 72-hour passages, sustained expansion through serial culture, maintenance of representative MSC phenotypes, and adipogenic, osteogenic, and chondrogenic differentiation potential.

Biofargo MSC Max Kit is suitable for laboratories seeking a consistent mesenchymal stem cell growth medium for research involving human umbilical cord MSCs, bone marrow MSCs, adipose-derived MSCs, MSC banking, differentiation studies, and cell manufacturing process development.

Recommended Culture Workflow

Prepare the complete MSC expansion medium by combining the Biofargo MSC Max basal medium with the supplied supplement using aseptic technique. Follow the laboratory’s validated preparation and handling procedures when mixing, aliquoting, and storing complete medium.

For routine passaging, use a gentle stem cell dissociation enzyme and seed the harvested human MSCs at approximately 8,000–10,000 cells/cm². Under the evaluated workflow, cells were cultured for approximately 72 hours per passage. Culture vessels did not require precoating under the recommended conditions.

Cell morphology, attachment, confluence, and proliferation rate should be monitored during culture. Seeding density, passage timing, dissociation conditions, and medium volume may require optimization for different donors, tissue sources, culture vessels, and experimental objectives.

Expansion Performance

Serial Passage of Human Mesenchymal Stem Cells

Biofargo MSC Max Medium was evaluated during ten consecutive passages of human MSC culture. Cells were seeded at 8,000 cells/cm² during passages 1–7 and at 10,000 cells/cm² during passages 8–10. Each passage was cultured for approximately 72 hours.

Passage	Seeding Density (cells/cm²)	Culture Time (hours)	Harvested Cells (×10⁶)	Expansion Fold
01	8,000	72	2.57	13.3
02	8,000	72	2.19	10.9
03	8,000	72	2.10	10.5
04	8,000	72	2.03	11.1
05	8,000	72	1.95	9.7
06	8,000	72	1.74	10.4
07	8,000	72	1.75	9.0
08	10,000	72	1.99	8.1
09	10,000	72	2.09	8.4
10	10,000	72	1.96	7.9

Across the evaluated ten-passage culture period, the measured expansion ranged from approximately 7.9-fold to 13.3-fold per passage. These data are representative of the tested cell source and experimental conditions and should be used as reference performance data rather than a guaranteed result for every MSC line.

MSC Phenotype and Multipotency

Phenotype Maintenance After Serial Culture

Human MSCs cultured with Biofargo MSC Max Medium were evaluated by flow cytometry after ten passages. The representative histograms demonstrate phenotype analysis following extended serial expansion in the serum-free culture system.

Marker expression and acceptance criteria should be established according to the laboratory’s validated MSC characterization panel, tissue source, donor characteristics, and intended research application.

Three-Lineage Differentiation Potential

Following continuous culture in Biofargo MSC Max Medium, the expanded MSCs were subjected to adipogenic, osteogenic, and chondrogenic induction. Representative staining results showed that the cultured cells retained the ability to undergo three-lineage mesenchymal differentiation under the evaluated conditions.

Why Choose Biofargo MSC Max Kit?

Serum- and platelet lysate-supplemented MSC media can provide effective growth, but differences between raw-material lots may affect proliferation, cell morphology, and downstream experimental consistency. These variables become increasingly important during long-term culture or when comparing MSCs from different donors and tissue sources.

Biofargo MSC Max Kit provides a serum-free and animal component-free human MSC culture system developed to reduce dependence on undefined serum components. The dedicated basal medium and supplement are optimized as a complete mesenchymal stem cell expansion medium for routine propagation, serial passage, and differentiation research.

The evaluated workflow combines a recommended seeding density of 8,000–10,000 cells/cm² with gentle enzymatic dissociation and no precoating requirement. This streamlined approach can help laboratories establish a more standardized human MSC culture process while retaining flexibility for donor-specific and application-specific optimization.

Important Culture Considerations

Performance may vary according to donor, tissue source, isolation method, passage history, initial cell quality, and culture vessel.
Confirm cell identity and MSC phenotype using an appropriate laboratory-validated characterization panel.
Optimize seeding density when transferring cells from serum-containing or platelet lysate-containing culture systems.
Avoid excessive dissociation time or harsh mechanical handling that may reduce cell attachment and viability.
Monitor cultures regularly and determine passage timing according to cell morphology and confluence rather than culture time alone.
Validate downstream differentiation, extracellular vesicle, or molecular analysis workflows using the specific MSC source and experimental design.
