Description

Lipoprotein-Depleted Fetal Bovine Serum (LPDS) is fetal bovine serum from which lipoproteins have been removed by density ultracentrifugation. Removing LDL, HDL and VLDL takes away the main external source of cholesterol and other lipids that cultured cells normally take up from serum, while keeping the serum proteins and growth-supporting factors.

Cells grown in LPDS must rely on their own cholesterol synthesis and lipoprotein uptake pathways. This makes LPDS the standard serum supplement for studying cholesterol biosynthesis, SREBP signaling, LDL receptor regulation and lipoprotein uptake. After ultracentrifugation, the serum protein concentration is adjusted to approximately 35 mg/mL with phosphate-buffered saline (PBS), and the product is sterile-filtered, aseptically packaged and frozen.

Features

Lipoproteins removed by ultracentrifugation — the reference method for preparing lipoprotein-deficient serum.
Serum proteins retained and adjusted to about 35 mg/mL, so cells stay viable and attached in lipid-depleted conditions.
Removes exogenous cholesterol, activating SREBP-2 and LDL receptor expression in cultured cells.
Lot-tested for depletion by agarose gel electrophoresis with lipid staining.
Three pack sizes (10, 50 and 100 mL) for pilot work through larger studies.

Applications

Cholesterol Biosynthesis: Study the mevalonate pathway and HMG-CoA reductase regulation under sterol-depleted conditions.
SREBP Signaling: Induce SREBP-2 processing and target gene expression.
LDL Receptor and PCSK9 Research: Up-regulate LDL receptor expression for LDL binding and uptake assays.
Lipoprotein Uptake: Measure uptake of labeled LDL, HDL or VLDL without competition from serum lipoproteins.
Drug Screening: Evaluate statins and other lipid-lowering compounds in sterol-depleted cells.
Lipid Metabolism in Cancer and Immunology: Examine how tumor and immune cells depend on exogenous lipids.

Storage & Shipping

Store at ≤ −20 °C. Avoid repeated freeze–thaw cycles.
Thaw at 2–8 °C or at room temperature, mix gently, and aliquot for single use.
Shipped on dry ice.

Protocol Overview

Thaw the serum at 2–8 °C overnight or at room temperature; avoid prolonged warming at 37 °C.
Mix gently by inversion and aliquot to avoid repeated freeze–thaw.
Replace regular FBS with LPDS in the culture medium, typically at 5–10% (v/v). The optimal concentration and exposure time depend on the cell type and assay.
For sterol-depletion protocols, culture cells in LPDS-containing medium for 16–24 hours before stimulation or analysis; statins can be added to further suppress endogenous cholesterol synthesis.

Frequently Asked Questions (FAQs)

Common questions about Lipoprotein-Depleted Fetal Bovine Serum (LPDS).

What is lipoprotein-depleted serum used for?

It is used to culture cells without exogenous lipoproteins, for studies of cholesterol synthesis, SREBP signaling, LDL receptor regulation, lipoprotein uptake and lipid-lowering drugs.

How are the lipoproteins removed?

Lipoproteins are removed by density ultracentrifugation, then the serum protein concentration is adjusted to about 35 mg/mL with PBS.

Can LPDS completely replace FBS in my medium?

Yes. LPDS is used in place of regular FBS, usually at 5–10%. Some cell types grow more slowly in lipid-depleted conditions, so check viability for your cells.

Is lipoprotein-depleted serum the same as delipidated or charcoal-stripped serum?

No. LPDS has lipoproteins removed by ultracentrifugation. Delipidated serum is solvent-extracted to remove lipids, and charcoal-stripped serum mainly removes hormones and other small lipophilic molecules.

How should LPDS be stored?

Store at −20 °C or below and aliquot after the first thaw to avoid repeated freeze–thaw cycles.

Can it be used for clinical or diagnostic purposes?

No. This product is for research use only and is not intended for human, diagnostic or therapeutic use.
